Gastric carcinoma is one of the common malignant tumors, and is locatedthe first place at incidence and death rate of all kinds of alimentary tracttumors in China. Occurrence of gastric carcinoma is a multiple factors,multiple steps process that involves the various mechanisms. At present, it isconsidered that pathogenesis of gastric carcinoma relates to integrinα5β1,CD151and heparanase of gastric tumor cell.To investigate the relationship between integrinα5β1, CD151, heparanaseand clinical pothology characteristics(differentiation, TNM staging, metastasisof lymph nodes, depth of invasion) of gastric carcinoma by detecting theexpression of them in normal and carcinomatous gastric tissue, for providingtheoretical basis to the study of the pathogenesis of gastric carcinoma and newtargets of clinical treatment.Objective:To reveal the expression of integrinα5β1, CD151and heparanase innormal and carcinomatous gastric tissue by immunohistochemisty staining andin situ hybridization histochemistry. To research the relationship amongintegrinα5β1, CD151and heparanase in gastric carcinoma through analysingexperimental data of them.Methods:To collect80cases of gastric carcinoma and50cases of normal gastrictissue(away the tumor more than10centimetres) which were surgicallyremoved and certified by pathology in many hospitals of Chengde, Tangshan,Baoding from October,2011to March,2012. Immunohistochemisty stainingwas adopted to detect the expression of integrinα5β1protein, CD151proteinand heparanase protein in normal and carcinomatous gastric tissue; the expression of integrinα5β1mRNA, CD151mRNA and heparanase mRNAwas detected by in situ hybridization histochemistry in normal andcarcinomatous gastric tissue. To analyse the experimental data combinedclinical pathology with statistics.Results:1.1The expression of integrinα5β1protein in carcinomatous gastrictissue was obviously higher than that in normal gastric tissue (P<0.05). Theexpression of CD151protein in carcinomatous gastric tissue was obviouslyhigher than that in normal gastric tissue (P<0.05). The expression ofheparanase protein in carcinomatous gastric tissue was obviously higher thanthat in normal gastric tissue (P<0.05).1.2.1The positive expression of integrinα5β1protein in gastriccarcinoma which were poorly differentiated was significantly higher than thatin well differentiated and moderately differentiated gastric carcinoma(P<0.05). The positive expression of integrinα5β1protein in gastric carcinomaof TNM stage Ⅲ and Ⅳ was significantly higher than that of TNM stageⅠand Ⅱ(P<0.05). The expression of integrinα5β1protein in gastric carcinomawith lymph node metastasis was significantly higher than that in gastriccarcinoma without lymph node metastasis (P<0.05). The positive expressionof integrinα5β1protein in gastric carcinoma of which invasion depth reachedserous membrane was significantly higher than that in gastric carcinoma ofwhich invasion depth did not reach serous membrane (P<0.05).1.2.2The positive expression of CD151protein in gastric carcinomawhich were poorly differentiated was significantly higher than that in welldifferentiated and moderately differentiated gastric carcinoma (P<0.05). Thepositive expression of CD151protein in gastric carcinoma of TNM stage Ⅲand Ⅳ was significantly higher than that of TNM stage Ⅰ and Ⅱ (P<0.05).The expression of CD151protein in gastric carcinoma with lymph nodemetastasis was significantly higher than that in gastric carcinoma withoutlymph node metastasis (P<0.05). The positive expression of CD151protein ingastric carcinoma of which invasion depth reached serous membrane was significantly higher than that in gastric carcinoma of which invasion depth didnot reach serous membrane (P<0.05).1.2.3The positive expression of heparanase protein in gastric carcinomawhich were poorly differentiated was significantly higher than that in welldifferentiated and moderately differentiated gastric carcinoma (P<0.05). Thepositive expression of heparanase protein in gastric carcinoma of TNM stageⅢ and Ⅳ was significantly higher than that of TNM stage Ⅰ and Ⅱ (P<0.05).The expression of heparanase protein in gastric carcinoma with lymph nodemetastasis was significantly higher than that in gastric carcinoma withoutlymph node metastasis (P<0.05). The positive expression of heparanaseprotein in gastric carcinoma of which invasion depth reached serousmembrane was significantly higher than that in gastric carcinoma of whichinvasion depth did not reach serous membrane (P<0.05).There were no relationships between integrinα5β1protein, CD151protein, heparanase protein and every other characteristics of gastriccarcinoma such as age, gender, type of pathology (P>0.05, P>0.05, P>0.05).2.1The expression of integrinα5β1mRNA in carcinomatous gastrictissue was obviously higher than that in normal gastric tissue (P<0.05). Theexpression of CD151mRNA in carcinomatous gastric tissue was obviouslyhigher than that in normal gastric tissue (P<0.05). The expression ofheparanase mRNA in carcinomatous gastric tissue was obviously higher thanthat in normal gastric tissue (P<0.05).2.2.1The positive expression of integrinα5β1mRNA in gastriccarcinoma which were poorly differentiated was significantly higher than thatin well differentiated and moderately differentiated gastric carcinoma(P<0.05). The positive expression of integrinα5β1mRNA in gastric carcinomaof TNM stage Ⅲ and Ⅳ was significantly higher than that of TNM stageⅠand Ⅱ (P<0.05). The expression of integrinα5β1mRNA in gastric carcinomawith lymph node metastasis was significantly higher than that in gastriccarcinoma without lymph node metastasis (P<0.05). The positive expressionof integrinα5β1mRNA in gastric carcinoma of which invasion depth reached serous membrane was significantly higher than that in gastric carcinoma ofwhich invasion depth did not reach serous membrane (P<0.05).2.2.2The positive expression of CD151mRNA in gastric carcinomawhich were poorly differentiated was significantly higher than that in welldifferentiated and moderately differentiated gastric carcinoma (P<0.05). The positive expression of heparanasemRNA in gastric carcinoma of which invasion depth reached serousmembrane was significantly higher than that in gastric carcinoma of whichinvasion depth did not reach serous membrane (P<0.05).There were no relationships between integrinα5β1mRNA, CD151mRNA, heparanase mRNA and every other characteristics of gastriccarcinoma such as age, gender, type of pathology (P>0.05, P>0.05, P>0.05).3.1The correlation coefficient of integrinα5β1protein and CD151protein is: r=0.428(P<0.05); The correlation coefficient of integrinα5β1protein and heparanase protein is: r=0.564(P<0.05); The correlationcoefficient of CD151protein and heparanase protein is: r=0.469(P<0.05). 3.2The correlation coefficient of integrinα5β1mRNA and CD151mRNA is: r=0.541(P<0.05); The correlation coefficient of integrinα5β1mRNA and heparanase mRNA is: r=0.557(P<0.05); The correlationcoefficient of CD151mRNA and heparanase mRNA is: r=0.594(P<0.05).Conclusions:1. Immunohistochemical positive signals of integrinα5β1protein, CD151protein and heparanase protein were significant differences between normalgastric tissue and gastric carcinoma, also were concerned with differentiationdegree, TNM staging, metastasis of lymph nodes, depth of invasion of gastriccarcinoma.2. Integrinα5β1mRNA, CD151mRNA and heparanase mRNA weresignificant differences between normal gastric tissue and gastric carcinoma,also were concerned with differentiation degree, TNM staging, metastasis oflymph nodes, depth of invasion of gastric carcinoma.3. The correlation of integrinα5β1and CD151was positive; Thecorrelation of integrinα5β1and HPA was positive; The correlation of CD151and HPA was positive; The high expression of integrinα5β1, CD151and HPAmay play positive cooperativity in the occurrence and development of gastriccarcinoma.
