Analysis The Expression Of NGF And VEGF In Human Breast Cancer Cell(MCF-7) Treated With Iodine-125 And Study Their Correlation

Ojective:To investigate the effect of the iodine-125 on expression of nerve growth factor and vascular endothelial growth factor in human breast cancer,and to explore correlation between NGF and VEGF,certificate the efficiency to therapy on breast cancer by implanting iodine-125 with the new view.Methods:Take the female nude mice of BLAB/c-nu/nu with MCF-7 tumor of breast cancer for animal models,and follow by the implant of the iodine-125 seeds of different group into the tumor mass. To implant iodine-125 seeds,the animal are divided into two groups(one group contains 24 nude mice),control group:to implant blank seeds,without iodine-125 radioelement(0mCi);experiment group:to implant iodine- 125 seeds(0.4mCi),and to observe the condition of tumor growth after implanting iodine-125.To study inhibitive effect to breast cancer,in 7th,14th,21th,28th day after implantation, we kill 12 nude mice(one group contains 6 nude mice),conserve tumors.detect the mRNA and protein expression levels of NGF and VEGF at different time-point by RT-PCR and immunohistochemistry,and undertake NGF and VEGF correlation analysis.Results:after implanted iodine-125 in tumor for 21 days,the expressions of NGF and VEG F in level of proteinum and mRNA in tumor tissue are significantly different,and they may be even lower for 28 days.there are significantly different compared with the controls(P＜0.05),NGF and VEGF expressions of tumor tissue are highly positive correlation(P＜0.05).Conclusions:The important molecular biology mechanism to restrain breast cancer with iodine-125 seeds is to cut down the expression of NGF and VEGF in tumor cells;expressions of NGF and VEGF are highly positive correlation in human breast cancer cells(MCF-7).
