Objective: To observe the inhibition effect of Curcumin for endometriosis(EMs) and its influence on vascular endothelial growth factor (VEGF) and microvessel density (MVD) in expressed eutopic and ectopic endometrium of experimental rats, exploring the pathogenesis of endometriosis and offering more experimental evidence for clinical use of Curcumin.Methods: An improving rat model of endometriosis was established. 48 Female virgin rats in estrus stage were conducted by autotransplantation of its own endometriosis, after 28 days eight rats were executed randomly so as to confirm with pathology that the rat model was made successfully. Remain rats were divided randomly into four groups, which three groups were intragastrically administrated with Curcumin(150mg/kg, 100mg/kg, 50mg/kg, individually), and the model group was administrated with dissolvent for Curcumin intragastrically. All the rats were treated everyday for 28 days continuously, examined by histology and immunohistochemical staining for MVD of eutopic endometrium and ectopic endometrium. VEGF levels were measured in eutopic endometrium and ectopic endometrium by Western blotting.Results:1. It was confirmed with pathology that the rat model was made successfully by autotransplantation of its own endometrium between the superficial and muscular layer of the abdomial wall.2. The cubic capacity of focal tissue in gross appearance was great in model group, and was diminuted significantly after treatment with Curcumin (P < 0.05), which effects are dose-dependent.3. MVD in the eutopic endometrium was higher in model group compared with that of normal endometrium(P > 0.05), and MVD in the ectopic endometrium was increased obviously which was decreased significantly after treatment with Curcumin (P<0.05), which effects are dose-dependent.4. The level of VEGF in the eutopic endometrium was higher in model group compared with that of normal endometrium(P > 0.05), and the expression of VEGF in the focal tissue was raised obviously which was significantly depressed after treatment with Curcumin (P < 0.05), which effects are dose-dependent. The correlation between MVD and VEGF was positively.Conclusion:1. The rat model of endometriosis with angiogenesis which reformed and conducted by autotransplantation of its own endometrium was successfully made.2.It was illustrated that the heterogeneity is existed between eutopic and ectopic endometrium on account of MVD and the expression of VEGF in the eutopic endometrium was not similar in model group compared with that of ectopic endometrium.3. Curcumin can decrease the cubic capacity of focal tissue and reduce the neovascularization, inhibit the formation and development of endometriosis.4. It can be deduced that VEGF was play a considerable role in endometriosis, but Curcumin can decrease the quantity of microvessel and the expression of protein VEGF in heterotopic endometrium of endometriosis rats.
