Objective To study the expressions of vascular endothelial growth factor(VEGF) and microvessel density(MVD)-CD34 in serum, hepatic alveolar hydatid tissue and liver tissue in inoculated alveolar hydatid gerbil model and explore their clinical significances. Methods Sixty health gerbils were randomly equally divided into three groups, a model group, a control group and a sham operation group. In model group each gerbil was given liver vaccination by opening their abdominal. Each gerbil in the model group was injected with alveolar echinococcus protoscolex of immature suspension mixing of turbid liquid 0.1m L, and each gerbil in the control group received a corresponding volume of phosphate buffered saline, then each gerbil in the sham operation group had direct suture abdominal liver puncture in open abdominal cavity after operation. Gerbils in each group were were sacrificed on day 120, after taking blood from their body. The alveolar hydatid tissue, surrounding liver tissue and distal liver tissue were collected from the model group and the normal liver tissue was collected from the control group and sham operation group, and the expressions of VEGF and MVD-CD34 were detected by immunohistochemistry staining, Western Blot and enzyme-linked immunosorbent assay(Elisa) analysis. Results Echioncoccus multilocularis hydatid tissues were observed over the liver and the liver of other groups were normal. The expressions of VEGF were observed in the different tissue: after infection and the positive cells of VEGF were yellow or brown yellow granules, located in the cytoplasmic. The scores of VEGF and the ratio of VEGF/ β-actin in the different parts were 5.13±1.81and1.12±0.05, 4.20±1.52 and 0.91±0.02, 2.46±1.06 and 0.28±0.02,2.20±0.77 and 0.30±0.02, 2.13±1.72 and 0.25±0.01. The expression of VEGF in different histological site showed a gradient change, in bubble hydatid tissues VEGF expressed the highest when compared with several other groups(P < 0.05). The expressions of CD34 were observed in the alveolar hydatid tissue and surrounding liver tissue after infection and the positive cells of CD34 was brown, mainly located in the cell membrane of endothelial cells. The scores of CD34 and the ratio of CD34/ β-actin in the different parts were 5.06±2.46 and 0.66±0.01,2.43±1.26 and 0.58±0.02,and the rest parts did not detect the expression of CD34. The expressions of CD34 were observed in the alveolar hydatid tissue is higher than any other tissues(P < 0.05). The expression quantity of VEGF of each group was:106.17±14.17 pg/m L, 38.13±21.36 pg/m L, 42.88±17.25 pg/m L;The expression quantity of CD34 of each group was: 1.47±0.28 ng/m L; 0.82±0.12 ng/m L; 0.72±0.16 ng/m L, group A compared with group B and group C, the difference had statistical significance(P<0.05). Conclusion There are signs of angiogenesis in gerbil alveolar hydatid tissues, Angiogenesis might be involved in infiltrated growth of alveococcus.
