| Objective:To study the change of Leptin, Testosterone (T), Luteinizing hormone(LH) , follicle-stimulating hormone (FSH) in the serum and the level of ob mRNA in ovary of polycystic ovarian (PCO) rat before after therapy With magnolol; to contrast with Metformin, and initially approach it's mechanism of action.Methods:Sixty female SD rats aged 8 weeks after feeding 4d were divided into control group (n=10), PCO model group (n=50), All rats were maken model besides control group. The control group were given common animal feeds and common water; The rats in the model groups were induced by Poretsky's method for PCO modeling, evaluating the effect of making modle after 22d.The modeling were divided into model group(n=10), treatment group(Magnolol) (n=10) and control group(metformin) (n=10) randomly. followed by administration of Sodium Chloride 1ml/100g/d or magnolol 5mg/100g/d or metformin 15mg/100g/d in respectively.After 21 d treatment, the rats were sacrificed and the level of Leptin,T,LH and FSH in the serum were detected by Elivision TM Plus. The ovary were taken down, the moiety were maken patho-section and another moiety were detected ob mRNA by Fluorescent Quantitation PCR.Results:1.Macroscopic observation found that the model making groups' ovarial surface were pale and the volume increasing, and small amounts corpus luteum, Observing by microscope that the ovarian follicle extend and oocyte or corona radiata lose, and the granular cell layers decreased to 2～3 layers and artially follicular atresia.These indicated the model making is successful(Fig 1 and 2).2.Compared with normal control group, The ovarian colour were red and the corpus luteum can be found of the treatment group, there are many corpus luteum and all ovum be found by microscope.The Saccular dilatation decreased obviously or abolition, granular cell layer increased, follicular fluid and ovarium mound can be found in dominant follicle (Fig 3 and 4).3. The model making groups rats' body weights was significantly higher than the nomal control group (P<0.05), the magnolol group and metformin group rats' body weights was significantly decreased than the model groups (P<0.05). It is no statistics defference VS the metformin group(P>0.05), these indicated magnolol could impove the PCO rats functional condition (List 1) .4. The Leptin and T and LH were increased obviously (P<0.05) in the serum of PCO group but that of FSH was decreased.In the magnolol treatment group, the Leptin, T and LH level declined(P<0.05), which were lower than the levels of control group.the results indicated that the magnolol could improved rats' serum hormone levels (List 2) .5.The level of ovary ob mRNA expression in model group significantly increased(P<0.05).compared with the model making groups, the medicine groups decreased obviously(P<0.05), also infra the positive control groups, But it is no statistics defference VS the metformin group(P>0.05), the results indicated that the magnolol could improved rats' ovary function (Fig 8) .Conclusions:therapeutic action is significant of magnolol to the modeling rats of PCO, That it can alter ovarian volume.colour.and pathology state, significantly decreased obesity rats' body weights of PCO and improve their functional condition; magnolol could decreased rats' serum hormone levels(Leptin,T,LH) and ovary ob mRNA expression, and the therapeutic effect is better than metformin.therapeutic action of magnolol to the modeling rats of PCO possible by cut down cholesterol and low density lipoprotein(LDL), and cut down the differentiation degree of fat cell, accordingly to cause the expression deceased of ob gene and leptin level decreased in serum. Because the serum leptin is intercommunication and interrestrict with other hormone, So serum T,LH also degrade. |
