Selaginella pulvinata ( hook et Grev ) Maxim,which is a traditional chinese herb, is widely spread in China. It's same genus plant Selaginella tamariscina has been adopted into Chinese pharmacopeia ( 2005 ), and more than 20 plants in Selaginella genus are used as medicine. Pharmacological investigation shows that they possess complex Pharmacological actions such as anti-inflammatory, anti-bacterial,anti-virus, immunity-stimulating, anti-tumor, analgesia, hypoglycemic effect, Spasmolysis and so on.To explore the medical effect compounds, we studied the whole herb systematically,determining content of amentoflavone from Selaginella pulvinata by HPLC. In our experiments, the material has been immersed in 70%EtOH. The concentrated extract was treated by centrifugal machine. Compound I was obtained from the deposit after re-silica gel CC. solution was treated by D101 macroporous adsorption resin, eluted by water, 30% EtOH, 70% EtOH, 95% EtOH separately.Compound II,III,IV and V were obtained from the concentrated solution of 30% EtOH through re-silica gel CC, Sephadex LH-20 CC and preparative HPLC. VI and VII were achieved from the concentrated solution of 70% EtOH through re-silica gel CC and Sephadex LH-20 CC. On the basis of spectroscopic value and comparison with the authentic literatures,their structures were identified as:β-sitosterol ( I ), adenosine ( II ), tamariscinoside A ( III ), syringaresinol-4,4'-0-bis-β-D-glucoside ( IV ), caffeic acid ( V ), amentoflavone ( VI ) , apigenin ( VII ). Among of them, compounds II, III, IV, V are obtained from Selaginella pulvinata Maxim. for the first time.In addition, amentoflavone is its main chemical constituent, its content can be used as comparison index of it's quality standard. Method of determining its content by RP-HPLC was sensitive, simple, exact was suitable for the determination of amentoflavone. In order to supply the reference for the study of quality standard,we determined content of amentoflavone in Selaginella pulvinata from five different areas.
