Objective: To explore the regulation and mechanism of action of low molecular weight heparin (LMWH) on cell proliferation of human stomach cancer cell line SGC-7901.Methods: The MTT method was employed to measure the growth inhibition rates at different concentrations of LMWH; the flow cytometer (FCM) was used to analyze the distribution of cell cycle and apoptosis; the semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) was used to assay the changes of p27 mRNA expression.Results: After treated with different concentration of LMWH in the range of 0.0625~1 mg/ml at 24hours and 48hours, the concentration of LMWH at 0.0625mg/ml had an inhibition rate of (18.66±10.97)% and (27.66±7.99)%, the concentration of LMWH at 1mg/ml had an inhibition rate of (44.20±10.64)% and (68.90±12.32)%, so LMWH could inhibit the proliferation of stomach cancer cells in dose and time-dependent manner. The concentration of LMWH at 0.0625 and 1mg/ml at 48 hours had an apoptosis rate of (0.84±0.55)% and (1.61±0.72)%. The distribution ratios of cells were significantly different compared with those of the controls (P<0.05). The number of G0/G1-phase cells increased and S-phase cells decreased. On RT-PCR, when the concentration of LMWH was 0.0625, 0.25 and 1mg/ml, the value of P27mRNA/β-actin was 0.44±0.03, 0.71±0.05 and 0.93±0.04, it was significantly different compared with those of the controls (P<0.05).Conclusion: LMWH has significantly inhibiting effects on the growth of SGC-7901 cell.The mechanism is probably related to its causing G0/G1 arrest and enhancing the expression of p27 mRNA.Thus the anti-proliferative action is achieved.
