PartⅠ:Objective:To sieve out the Human Bladder Cancer Cell T24 in Vitro which has high capability of invasion and metastasis from cell line T24 and investigate the expression of uPA system in Bladder Cancer Cell and its effect in invasion and metastasis.Methods:Sieve out Bladder Cancer Cell T24-C from T24 by Transwell Cabin,and then test the absorbance(A)of two cell lines in 0.5h,1h and 2h by method of MTT, meanwhile test numbers of invasion and metastasis between this two cell lines in 48h;the expression of uPA mRNA in cultured T24 cells and T24-C cells were examined by RT-PCR.Results:Compared with T24 and T24-C,the A(absorbance)in 0.5h has no statistical significance(P=0.092);The A in 1h and 2h has statistical significance(P<0.05).The numbers of invasion and metastasis between this two cell lines were different and the result has statistical significance(P<0.01).The expression of uPA mRNA between this two cell lines is different and the result has statistical significance(P<0.01).Conclusions:In vitro,it is feasible to sieving out Bladder Cancer Cell which has high capability of invasion and metastasis,the uPA system plays an important role in invasion and metastasis of bladde cancer.PartⅡ:Objective:To study the effect of Amilorde which is the synthetic inhibitor of urokinase plasminogen activator to the adherent,and the invasion and metastasis of cell line T24 in vitro. Methods:Test the A of cell line T24-C with 200μmol/L amilorde in 0.5h,1h and 2h by method of MTT,and then determine the inhibitory action of different concentration amilorde(0μmol/L,100μmol/L,150μmol/L,200μmol/L,250μmol/L)to invasion and metastasis of T24-C by Transwell Cabin.The expression of uPA mRNA in T24-C cells was examined by RT-PCR.Results:Between control and different experimental groups,the A of adherent cells in 0.5h,1h and 2h has statistical significance(P<0.01),meanwhile the quantity of cell which can breakthrough the cell envelope has statistical significance(P<0.01).The expression of the four groups uPA mRNA was decreased compared with control group.In the experimental groups,the effect between 200μmol/L and 250μmol/L has not statistical difference(P=0.781).Conclusions:Amilorde can significantly inhibit adherent,invasion and metastasis of T24-C.In effective concentration,and the inhibitory action is stronger than the concentration step up.The concentration of 200μmol/L is the most suitable group.
