Objiect:to investigate the roles of matrix metalloproteinase-9 and transforming growth factor-β1 in hypertensive vascular remodeling and intervention with atorvastatinMethods:Using two kidney one clip to establish renal hypertension rat model,24 rats were randomly divided into the control group(8),hypertension group(8),atorvastatin group (8).After four weekend,we began to gavage eight weeks with atorvastatin.Systolic blood pressure(SBP) was measured.Vascular structural changes was observed in light microscopy and the ratio of media thickness to lumen diameter(M/L) of thoracic aortic was calculated.Matrix metalloproteinase-9(MMP-9) and transforming growth factor -β1(TGF-β1) were evaluated by immunohistochemical staining to explore their expression.Results:Compared with the control group,SBP,M/L were significantly increased in hypertension group.MMP-9 expression was significantly enhanced,and TGF-β1 expression level was significantly lower(P<0.01).Atorvastatin group than hypertension group p M/L was significantly lower(P<0.05).MMP-9 expression levels were obviously lower,and the level of TGF-β1 expression was obviously increased.Conclusion:MMP-9 may be related to the formation of hypertensive vascular remodeling. TGF-β1 may be the protective factor in hypertensive vascular remodeling.Atorv astatin can inhibit the formation of myocardial fibrosis in rats,which may be related to decreased the expression of MMP-9 and enhanced the expression of TGF-β1 are closely linked.
