Objective:To investigate the relationship between adiponectin gene single nucleotide polymorphism (SNP) and type 2 diabetes and diabetic macroangiopathy in Shanxi Han population; To investigate the relationship between adiponectin receptor 1 gene(ADIPOR1) single nucleotide polymorphism(SNP) and type 2 diabetes in the Chinese hans of Shanxi.Methods:The genotypes of 276G/T of adiponectin were determined by polymerase chain reaction–restriction fragment length polymorphism (PCR-RFLP) in 210 persons of Shanxi Han nationality including 76 normal control subjects, 66 type 2 diabetic patients and 68 type 2 diabetes with macroangiopathy. Height, weight, body mass index (BMI), waist to hip ratio (WHR), systolic blood pressure, diastolic blood pressure, the levels of serum insulin and C-peptide, plasma glucose level and insulin resistance index were also assayed. The genotypes of -102T/G of ADIPOR1 were detected by polymerase chain reaction with sequence specific primers(PCR-SSP) in 50 type 2 diabetes and 50 normal controls. Their clinical parameters were also examined.Results:1(1)There was significant difference in the distribution of genotypes and allele frequencies of the SNP276 polymorphism between normal control group and type 2 diabetes (P<0.05). The frequencies of G allele in type 2 diabetes were significantly higher than that in normal control group(P<0.01). The risk of type 2 diabetes was increased in non-TT(GG+TT)genotype compared to TT genotype. There was no statistical difference in the distribution of genotypes and allele frequencies of the SNP276 polymorphism between type 2 diabetes with macroangiopathy and type 2 diabetes without macroangiopathy (P>0.05). (2) The BMI, systolic blood pressure, fasting plasma insulin, HOMA-IR and fasting plasma glucose were significantly higher in GG or GT genotype than in TT genotype in normal control subjects. The systolic blood pressure, fasting plasma insulin and HOMA-IR were significantly higher in GG or GT genotype than in TT genotype in type 2 diabetes without macroangiopathy. The HOMA-IR was significantly higher in GG or GT genotype than in TT genotype in type 2 diabetes with macroangiopathy. 2(1)There was no statistical difference in the distribution of genotypes and allele frequencies between type 2 diabetes and normal controls(P=0.681,P=0.451).(2) Fasting plasma insulin(P=0.000),HOMA-IR(P=0.000) and fasting plasma glucose (P=0.000)was significantly lower in G allele carriers than that in TT genotype in normal controls. (3)Fasting plasma insulin(P=0.000),HOMA-IR(P=0.002) and fasting plasma glucose(P=0.000) were significantly lower in G allele carriers than in TT genotype in type 2 diabetes.Conclusion:(1) Adiponectin gene SNP276 polymorphism was probably associated with type 2 diabetes in Shanxi Han population, but was not associated with type 2 diabetes with macroangiopathy. The G allele was a risk factor of insulin resistance and type 2 diabetes. (2) ADIPOR1 -102T/G polymorphism was probably associated with glucose metabolism and insulin resistance, but was not associated with type 2 diabetes in Shanxi Han population.
