BackgroundHuman hepatocellular carcinoma (HCC) represents a major health problem worldwide. It is the fifth most common malignancy and the third leading cause of cancer related mortality worldwide. Although the treatment of liver cancer is multidisciplinary, involves surgeons, radiologists, oncologists as well as hepatologists to ensure the best therapeutic results, each method has its contraindication and limit, given that there is a paucity of effective treatment optional available for HCC.Traditional Chinese medicine plays an important role in oncology treatment in China, herbs have been proved to have anti-tumor effect both by clinical practice and empirical study, and testified to be beneficial for longer survival and better life. Actinidia valvata Dunn exhibited anti-tumor and anti-inflammatory effects, and had been used widely for hepatoma, lung carcinoma and myeloma therapy for a long time. It is abundant in amino acids and inorganic elements that people need. Anti-tumor effects of Actinidia valvata Dunn had been investigated for the past five years, its injection had been confirmed to have a significant anti-tumor effect, significantly restrain the growth of primarily hypodermal tumor with no cell-mediate immunity inhibitive effects in vivo. We tried to find out useful chemical constituents from Actinidia valvata Dunn for further study, observed inhibitive effects on proliferation, adhesion, invasion and migration of hepatoma cell lines in vitro. Furthermore, anti-tumor and anti-metastasis effects in vivo were also investigated.ObjectiveObserve total saponin from root of Actinidia valvata Dunn (TSAVD) effect on proliferation, mobility, adhesion, invasion and migration of HCC lines in vitro by using MTT, wound healing, adhesion, invasion and migration assay; observe TSAVD effects on mouse with primarily hepatoma by establishing hypodermal model and metastasis hepatoma by establishing experimental pulmonary metastasis tumor model.MethodPartⅠEffects of TSAVD on HCC cell lines in vitro1 MTT assay was used to observe TSAVD effect on proliferation of HCC cell lines. Twohepatoma cell lines were chosen for further study, a suitable concentration range was also identified.2 Two HCC cell lines were cultured and treated with 400, 200, 100, 50μg/ml TSAVDrespectively for 24 hours to observe medicine effect on cell mobility, adhesion ability. Theinvasion and migration of HCC cell lines were examined in transwell chamber with 200,100, 50μg/ml TSAVD for 24 hours, the number of migrate cells were observed, inhibtiveratio in each assay was calculated.PartⅡEffects of TSAVD on hepatoma growth and metastasis in vivo1 Two differrent concentration of TSAVD injections were given to mouse with primarily hypodermal hepatoma. The weight, tumor weight, inhibitive ratio were calculated. The MVD, VEGF, bFGF expressions were also compared between different groups.2 Two differrent concentration of TSAVD injections were given to experimental distract hepatoma mouse. The number of pulmonary metastasis tumor was counted, the pathology changes of lung and other organs were also observed, spleen index, blood cells were also calculated to know whether it has secondary effect.ResultPartⅠEffects of TSAVD on HCC cell lines in vitro1 TSAVD was shown to have inhibitive effects on proliferation of HCC lines, can best inhibit BEL-7402 cell in a concentration dependent manner between concentration from 1.5 to 0.5 mg/ml. The inhibitive effect of TSAVD on the MHCC-97-H cell is weak with the highest inhibitive ratio of 36% at 1.5 mg/ml in 24 hours.2 TSAVD also can restrain mobility, adhesion, invasion and migration ability of HCC cell lines in wound healing, adhesion, invasion and migration assays.PartⅡEffects of TSAVD on hepatoma growth and metastasis in vivo1 TSAVD can significantly inhibit the growth of primarily hypodermal hepatoma. Inhibit the expression of MVD, VEGF, bFGF maybe one of the reasons.2 TSAVD can restrain hepatoma metastasis, accelerate the necrotize of tumor.ConclusionTSAVD can inhibit proliferation, mobility, adhesion, invasion and migration ability of HCC cell lines. TSAVD can inhibit the growth and metastasis of hepatoma.
