Objective:To optimize the extraction process for total saponins from Radix Ranunculi Ternati. Effects of RRTS on proliferation, Invasion and metastasis of hepatocarcinoma cell line HCCLM3 and MHCC97-H were studied so as to illustrate the further theoretical evidence of which as an anti-tumor drug may be provided.Methods: Single factor experiment and orthogonal array design methods were used to optimize the extraction process. HCCLM3 and MHCC97‐H of high metastatic hepatocellular carcinoma cell lines were used as the study object to detect the impact of concentrations of the saponins on the growth of liver cancer cells by MTT.The effects of RRTS on invasiveness and motoricity of cells were observed by invasion and migration assays. The model of nude mice bearing tumor were established by inoculating HCCLM3 cell and observe the tumor growth and metastasis by intragastric method of RRTS.Results: The optimal extracting condition was as follow: 85% alcohol acted as solvent,ultrasonic extraction time was 2.5 h,the ratio of material and solvent was 1∶5 and extracted 3 times, The yield can reach 0.895%. The cell survival rate of the saponins treatment group reduced compared with that of the control group.This effect becomes more obvious with the increasing concentration of the drug. The cell density reduced after the treatment of the saponins through the observation under the Phase‐contrast microscope.RRTS has inhibitory effects on invasion and metastasis of human liver cancer HCCLM3 and MHCC97‐H cells in vitro. The relative size of nude mice tumors and tumors weight in RRTS group are smaller than blank group,the difference is statistically significant(P<0.01). The metastasis rate of RRTS group are lower than the blank group.Conclution: RRTS can inhibit proliferation and reduce hepatocarcinoma cells invasion and motility ability.
