The Experiment Study On The Effect Of JianXin Granule In Chronic Heart Failure (CHF) In Rats

Objective: To investigate the mechanism of the JianXin Granule(JG) in treating chronic heart failure (CHF) by observing it'pharmacological effects in heart failure spontaneously hypertensive rats (SHR) and comparing different effects among groups of JG and captopril.Methods: 50 15-week-old spontaneously hypertensive rats (SHR) as a test object were randomly divided into control group, the low-dose group, the middle-dose group ,the high-dose group and Cap group (captopril),10rats each group. After 8 weeks'treatment, blood parameters (LVSP, LVEDP,±dp/dtmax) and neuroendocrine indicators (AngⅡ, BNP, ET-1, ALD)were respectively observed, , myocardial hypertrophy indicators (HW / BW, LVW / BW) were calculated. The morphological and structural changes of myocardial cells in left ventricle of SHRs were observed with light microscope. Then we will approach the methanisim of JG in chronic Heart Failure after the statistical analysis.Results:1. In high and middle groups ,the LVSP,±dp/dtmax were increased (p <0.05, p<0.01)and LVEDP decreased (p <0.05) Showing that JG can speed up myocardial contractility and improve the diastolic function2. In the groups of JG , the plasma angiotensin (AngⅡ), brain natriuretic peptide(BNP),aldosterone(ALD),endothelin(ET-1)levels were significantly reduced (p<0.05,P<0.01).there is no difference among the high-dose group , the middle-dose group and cap group (P> 0.05),which could Show that the JG can improve the level of neuroendocrine indicators in SHR.3. HWI (HW / BW) and LVMI (LVW / BW) in various dose groups of JG decreased more significantly than that in control group (P <0.01). there was no significant difference (p>0.05) among the middle-dose gronp , high-dose group and the cap group (captopril).That was to say, JG could improve cardiac index and the index of ventricular hypertrophy.4. The result under light microscope were: SHR myocardial cells in the high-dose group myocardial cell morphology most normal phenomenon of no significant degeneration and necrosis, suggesting that JG can inhibit cell hypertrophy and myocardial necrosis to enhance myocardial contractility.Conclusion: JianXin Granule (JG )can significantly enhance the myocardial contractile function to improve hemodynamic abnormalities. JG can also reduce significantly the levels of AngⅡand ALD in SHR, inhibit the activation of RAAS, regulate neuroendocrine levels , prevent ventricular remodeling. JG is the effective Chinese medicine to prevent and treat CHF with multi-system, multi-target, multi-directional comprehensive regulation.
