PURPOSES:To investigate the expression of NADPH oxidase and NF–κB in placenta during the course of preeclampsia, to explore the potential role of NADPH oxidase and NF–κB as well as their relationship during the course of preeclampsia .METHODS: 25 preeclampsic women were divided into mild preeclampsia group (n=10) and severe preeclampsia group(n=15). The normal control group were 25 healthy normotensive pregnancy. The expression of NADPH oxidase and NF–κB in placenta was qualitative and semiquantitative analysised with immunohistochemistry and reverse transcription polymerase chain reaction(RT–PCR)RESULTS:1. Immunohistochemistrys showed that both NADPH oxidase gp91phox and NF–κB p65 were mainly located in placental syncytiotrophoblasts, in vascular endothelial cells and a few in the stromal cells.2. Immunohistochemistrys demonstrated the expression of NADPH oxidase gp91phox in placenta of preeclampsia were stronger than that of control group(p<0.05),and there were stronger expression in severe preeclampsia than that in mild preeclampsia(p<0.05).3 . Immunohistochemistrys demonstrated the expression of NF–κB p65 were stronger in placenta of preeclampsia than that of control group(p<0.05),and there were stronger expression in severe preeclampsia than that in mild preeclampsia(p<0.05).4. There was correlation between the activity of NF–κB and the expression of NADPH oxidase ,the correlation coefficient was 0.822.5. RT–PCR showed that the expression of NADPH oxidase gp91phox mRNA were in parallel with what immunohistochemistrys showed.6. RT–PCR showed that the expression of NF–κB p65 mRNA in placenta of preeclampsia were not stronger than that that of control group(p>0.05)..CONCLUSION : In human placenta of preeclampsia showed increased expression of NADPH oxidase and activity of NF–κB. It suggests that NADPH oxidase and NF–κB participate in the pathogenesis of preeclampsia. Increasing NADPH oxidase in placenta induces NF–κB activity ,which leads to futher pathophysiological changes.
