This thesis is composed of three parts. Preface includes the review of chemical constituent and pharmacology of Ranunculus japonicus. PartⅠcovers our study on antiinflammatory and analgesic effect of TGOR. On partⅡ, this thesis reports study of total glycosides of Ranunculus japonicus(TGOR) on inhibitory effect of myocardial hypertrophy effect and the effects of frog heart and isolated thoracic aortic ring of rats. Finally the thesis introduces chemical constituent of Ranunculus japonicus.PartⅠStudy on antiinflammatory and analgesic effect of total glycosides of Ranunculus japonicusRanunculus japonicus distributes in most regions of China. It has several activities,such as antiinflammatory, antibiosis ,antitumor and so on. In folk, the whole plant was generally used for the therapy of many diseases: jaundice, analgesia, phymatiosis, hepatitis, et al. So far there is rare report on chemical and pharmacological study on Ranunculus japonicus in and abroad.Classic animal inflammation and pain models were applied to detect antiinflammatory and analgesic effect of TGOR. Results showed that 200mg/kg, 400mg/kg and 800mg/kg TGOR notably inhibited dimethyl benzene-induced ala auris swelling in mice,with dose dependence.200mg/kg and 400mg/kg TGOR significantly inhibited carrageen-induced feet swelling in rats.But 100mg/kg TGOR had no effect on the model,vs saline(p<0.05). 200mg/kg, 400mg/kg and 800mg/kg TGOR notably depressed pain response-induced by hot stimulation(with dose dependence) or ethanoic acid.The data mentioned indicated that TGOR possesed antiinflammatory and analgesia activity.PartⅡStudy on pharmacological effects of total glycosides of Ranunculus japonicus in vitroIn the thesis,the sulforhodamine B colorimetric assay were used to observe the effects of 20 kinds of traditional Chinese medicine extract on angiotensinⅡ-induced hypertrophic cadiocyte. It was detected that 50μg/ml TGOR underlied total protein level of angiotensinⅡ-induced hypertrophic cadiocyte. At the same time,the role of TGOR on flog heart perfusion and isolated thoracic aortic ring of rats was furtherly observed. Results show that 1μg/ml and 10μg/ml TGOR was able to underlied elementary contraction force level of isolated flog hearts and prevented myocardial contractility from rising in response to isoprenaline,but did not affect heart rate.1mg/ml or 2mg/ml TGOR had no effect on the elementary tension of arterial circle or isoprenaline-induced vasorelaxation.But it obviously inhibited noradrenaline-induced vasoconstriction.PartⅢChemical constituents of Ranunculus japonicusTo understand its substance foundation of effects and explore lead compound from Chinese materia medica, we studied chemical constituents of Ranunculus japonicus. Four compounds: anemonin(Ⅰ), ranunculinin(Ⅱ) , isoranunculinin(Ⅲ) and dihydroranunculinin (Ⅳ) were isolated from the polar part of Ranunculus japonicus. Among the compounds, isoranunculinin (Ⅲ) is a new compound, and dihydroranunculinin (Ⅳ) is isolated from the natural source for the first time.
