Objective: To discussion the effect and mechanism of total glycosides of ranunculus japoniucus on renal hypertensive rats.Method: We established renal hypertensive rats by used two kidney one clip (2K1C) and observed the effect of different doses of total glycosides of ranunculus japoniucus on rats'blood pressure, serum nitric oxide (NO), and the angiotensinⅡ(AngⅡ) in plasma and tissue. We also stained aortic of left ventricular and chest aortic by used Masson staining and observed the reconstruction of cardiovascular.Results: (1) The high, medium, low doses of total glycosides of ranunculus japoniucus can reduce the blood pressure of rats in varying degrees (P<0.05) . (2) The different doses of total glycosides of ranunculus japoniucus can significantly increase the oxide(NO) level in rats serum(P<0.01), but it show no significant effect on the content of angiotensinⅡ(AngⅡ)(P>0.05).(3) The different doses of total glycosides of ranunculus japoniucus can inhibition the abnormal proliferation of collagen fibers in left ventricular and chest aortic. It can improve the reconstruction of cardiovascular.Conclusion: The total glycosides of ranunculus japoniucus can improve and reduce the blood pressure of renal hypertensive rats.and it also can improve the reconstruction of cardiovascular in varying degrees. The mechanism that reduce the blood press may be related about the effect of increase the rats'NO level. But not AngⅡlevel. Therefore the mechanism of reduce the blood press may be not carried out by one single target. Objective: To observed the effect of total glycosides of ranunculus japoniucus on calcium ions level in vascular smooth muscle cells.Method: We used a new generations of Ca2+ fluorescent probe (Fluo-3/AM) to mark calcium in vascular smooth muscle cells which we cultured. Observed the fluorescence imaging by use inverted fluorescence microscope and measured the fluorescence intensity of calcium in vascular smooth muscle cells by use fluorescence spectrophotometer. To observed the effect of total glycosides of ranunculus japoniucus on calcium ions level in vascular smooth muscle cells of rats.Results: The angiotensinⅡcan significantly promote the calcium ions in extracellular inner flow. The different doses of total glycosides of ranunculus japoniucus can reduce the calcium ions level in cells which mediated by angiotensinⅡ. The results is related the doses of total glycosides of ranunculus japoniucus.Conclusion: Total glycosides of ranunculus japoniucus can antagonize the effect of AngⅡwhich can significantly promote the calcium ions in extracellular inner flowpartⅡThe effective part of total glycosides of ranunculus japoniucusObjective: To study the effect part of total glycosides of ranunculus japoniucus on vasodilatation in vitro.Method: We recorded the tension of thoracic aortic in vitro and observed the inhibition effect of different parts of total glycosides of ranunculus japoniucus on contraction of thoracic aortic which mediated by norepinephrine (NE) and KCI. As the Rutin to the standard and use UV spectrophotometer to measure the total flavonoid content in different parts of total glycosides of ranunculus japoniucus. Make the correlation regression of the flavonoid content as abscissa and the inhibition rates as vertical coordinate by Logistic and linear statistics.Results: (1) The different alcohol part of total glycosides of ranunculus japoniucus can inhibition the tension increased of thoracic aortic in varying degrees which mediated by norepinephrine and KCI (P<0.05).(2) Full-wavelength scanning to determine the maximum absorption wavelength is 358nm. The relative standard deviation precision of experiments and stability experiment RSD is <1.00%. The average recovery is in the range of 95%~105%. the relative standard deviation precision RSD<1.00%.(3)In the case of the contraction of thoracic aortic induced NE, Make the Logistic correlation regression of the flavonoid content and the inhibition rates, the correlation coefficient of 0.9559. and shows good correlation ;In the case of the contraction of thoracic aortic induced KCI, Make the Logistic correlation regression of the flavonoid content and the inhibition rates,the correlation coefficient of 0.9394.and shows good correlation.Conclusion: The flavonoid in total glycosides of ranunculus japoniucus glycosides might be the active components that inhibit the contraction of vascular which mediated by NE or KCI.
