Objective We compared effects of dragon's blood coprecipitates from different habitats on blood and cardiology majorly.Methods In the comparative studies of these coprecipitates on blood,we selected clotting time and suppressing effect of thrombotic formation as pharmacodynamic indexs,compared the different effects on the clotting time of normal mice,on the living time of congestive mice, on the platelet aggregate function of SD rats of blood stasis and hemorheological parameters. We also viewed their actions on plasma recalcification time in vitro.In the comparative studies of these coprecipitates on cardiology, we determined the possible mechanisms and principal active constituents of coprecipitates on the myocardial contractility and the heart rates both of normal isolated frogs'hearts and of ones pretreated by ISO. Meanwhile we observe preliminarily the antagonism of dragon's blood coprecipitates and simple substances, such as Loureirin A,Loureirin B and Dracorhodin Perochlorate, on the contraction of isolated rat thoracic aorta rings induced by KCl,noredrenaline(NE) and phenephrine(PE) respectively.Results1. We get clotting time by capillary tube method. Coprecipitates of dragon's blood-PEG6000-Poloxamer188 at high,middle and low dose show inhibitory actions on blood clotting of normal mice.Compared with blank group, all medication administration teams have significant differences(P<0.001). Coprecipitates of Yinni both 1.0g·Kg-1 group and 2.0g·Kg-1 group has the most powerful effect on prolonging clotting time. Di'aoxinxue Kang,Guangxi 2.0g·Kg-1,Yinni 1.0g·Kg-1 and 2.0g·Kg-1 can prolonging clotting time obviously, there are no statistical difference among those four groups(α=0.05),and the same to the others(α=0.05). It seems that both Guangxi and Yinni dragon's blood were dose-dependent. Coprecipitates of Yunnan 2.0g·Kg-1 group,Longwang 2.0g·Kg-1 group,Cambodia 2.0g·Kg-1 group and composite salvia miltiorrhiza tablets have less powerful effect on prolonging clotting time.2. Compared with model control group, all Coprecipitates,composite salvia miltiorrhiza tablets and Di'aoxinxue Kang improve the living time of congestive mice induced by collagen protein and epinephrine hydrochloride obviously(p<0.01). Guangxi 0.5g·Kg-1 group,Yinni 0.5g·Kg-1 group,Yunnan 2.0g·Kg-1 group have similar powerful effectiveness with composite salvia miltiorrhiza tablets and Di'aoxinxue Kang relatively,and there are no significant difference among them. Longwang 2.0g·Kg-1 group and Cambodia 2.0g·Kg-1 group have lower effect than composite salvia miltiorrhiza tablets and Di'aoxinxue Kang. It seems that Guangxi and Yunnan dragon's blood were dose-independent, and there are no significant differences between groups at high,middle and low dosage(p>0.05).3. There are significant differences among medication administration groups in recalcification time of blood plasma(P<0.05). Compared with blank group Guangxi dragon's blood flavones shorten recalcification time markedly(P<0.05), but the others have no statistical significant differences(P>0.05). Flavonoids maybe is one of principal constituents of dragon's blood which have some effect on blood.4 . Epinephrine hydrochloride combined with ice bath can increase platelets's maximum agglutination induced by ADP in ten minutes. Compared with model control group all medication administration groups have significant differences on platelets's maximum agglutination induced by ADP in ten minutes(P<0.05).It can be supposed that platelets aggregation of all medication administration groups was restrained markedly.Yinni 0.5g·Kg-1 group and Yinni1.0g·Kg-1 group,Guangxi 0.5g·Kg-1 group and 1.0g·Kg-1 group, gingko, these five groups have the more powerful action on the inhibition of platelets aggregation(comparing with model control group,P﹤0.01), and there are no significant differences among them(P﹥0.05). Salvia miltiorrhiza injection has the most poweful action on the inhibition of platelet aggregation. It seems that the effect of Guangxi dragon's blood on the inhibition of platelet aggregation was dose-dependent,and Yinni dragon's blood was dose-independent.5. We got the model of SD rats with blood stasis by epinephrine hydrochloride combined with ice bath. There is significant difference of model control group in specific viscosity under hypershear of whole blood and plasma of SD rats,compared with blank group and medication administration groups (P﹤0.001). Compared with model control group, all medication administration groups can lower specific viscosity under hypershear of whole blood and plasma of SD rats with blood stasis obviously(P﹤0.05),but there are significant difference compared with blank group(P﹤0.05). Guangxi 0.5g·Kg-1 group and gingko have poor effect on hemorheology, and have significant differences compared with others(P﹤0.05).Guangxi 1.0g·Kg-1 group and Yinni 0.5g·Kg-1 group have powerful effect on improving hemorheology, and have no significant differences compared with Di'ao xinxue Kang and Salvia miltiorrhiza injection(P>0.05). Yinni 1.0g·Kg-1 group have the most powerful effect on hemorheology, and have significant difference compared with others(P < 0.01). It seems that both Guangxi and Yinni dragon's blood were dose-dependent.6. Coprecipitate of Yunnan,Longwang and Yinni dragon's blood can lower myocardial contraction force induced by isopropylarterenol temporarily at 0.01mg/mL,and it seems that their effectiveness were dependent on their concentration. Coprecipitates of Dragon's blood from different habitates can inhibited markedly the contraction of aortic rings induced by NE and KCl at 0.1mg·mL-1 and 1mg·mL-1, and there are different effect on the aortic rings induced by PE. Yunnan and Longwang dragon's blood groups at 1 mg·mL-1 have strong inhibition on the contraction of aortic rings induced by NE. It seems that all kinds of Dragon's blood were dose-dependent of their inhibition effect on the contraction of aortic rings induced by NE. Jianlong dragon's blood groups at 1 mg·mL-1 have strong inhibition on the contraction of aortic rings induced by PE. Loureirin A,loureirin B and Dracorhodin Perochlorate can inhibit remarkably the contraction of isolated rat thoracic aorta rings induced by NE,KCl and PE.These studies and results mentioned above are mostly our exploratory studies, and they have not reported before. From these results, we can know, though domestic-made dragon's bloods and abroad ones have great differences in contents of primary reconstituents fairly, both of them have some similar pharmacological actions on blood and cardiology. They have latent value as medicine of prevention and cure on hematological and cardiovascular diseases, and can be used as substitutes between each other. The further researches are needed,such as isolating composition,approaching mechanisms and so on.
