| ObjectiveA continuous extraction method of Chinese traditional medicine was evaluated, with free total anthraquinone derivatives in Chinese traditional rhubarb and its compound prescription,and Angelica volatile oil inβ-CD inclusion compound extracted via liquid-liquid continuous extraction device.MethodsLiquid-liquid continuous extraction of free total anthraquinone derivatives from Chinese traditional rhubarb was compared with 3 common extraction methods,with content of anthraquinone determinated by UV.And the optimum novel extraction process was selected with the L9(43)orthogonal design content of anthraquinone derivative as index.Then by the novel extraction method,the extraction process of a compound prescription was evaluated,and a new assay of Angelica volatile oil inβ-CD inclusion compound was established.ResultsThe content of free total anthraquinone derivatives novel extraction was 2-3 folds of 3 common ones',RSD=1.28%in the former and RSD=5.40%in the latter.The optimum extraction condition of free anthraquinone derivatives from Chinese traditional rhubarb was:Adding 10-fold 2.5molv.mL-1sulfuric and continuously extracting for 5 hours.Through the novel extraction method,the optimum extraction of the compound prescription was obtained:Adding 7,5,5-fold times of 60%alcohol and extracting for 3 times,20min per time.The difference between new determination of content of Angelica volatile oil inβ-CD inclusion compound and the one of wet distillation was statistically meaningful due to higher 3.5%content of Angelica volatile oil and lower 2.9% of RSD.ConclusionThe novel continuous extraction with easy operation,high efficiency and reliable result was generalized not only in manufacture but also in quality control. |
