| Traditional Chinese medicine is the precious historical heritage of China. It is widely welcomed for its unique efficacy and minor side effects. Sample preparation is the pivotal process in the analysis of traditional Chinese medicine. In recent years, simple, efficient, rapid and environment friendly sample preparation becomes hot research topic. New preparation and determination methods for the analysis of anthraquinone derivatives in medicine and body fluid were studied in this thesis. The thesis has 4 chapters and the main results are as follows:In chapter 1:The principle and application of commonly used extraction methods are summarized, such as solid phase extraction, solid phase micro-extraction, supercritical fluid extraction, microwave-assisted extraction, liquid phase micro-extraction, dispersive liquid-liquid micro-extraction and cloud-point extraction. There is also a summary on the plant source, efficacy and extraction separation and quantitative analysis of rhubarb and semen cassiae.In chapter 2:A new micelle extraction-HPLC method has been developed and validated for the determination of anthraquinone derivatives in San-Huang tablet. Non-ionic surfactant Genapol X-080 was chosen as the extraction solvent. Some important parameters, such as the concentration of Genapol X-080 solution, liquid-solid ratio (mL·g-1) and ultrasonic-extraction time were investigated and optimized. Under the optimum conditions, five brands of San-Huang tablets were determined. The results demonstrate that this method is safe, accurate, reproducible and feasible, and could be used for the determination of anthraquinone derivatives in San-Huang tablets.In chapter 3:This study investigated the use of cloud point extraction method coupled with HPLC for the determination of anthraquinones from Chang-Qing Tea. Some important parameters, such as the concentration of Genapol X-080, liquid/solid ratio, extraction time, temperature and time for incubation and the concentration of NaCl were investigated and optimized. The LOD of this method is between 0.55ng·mL-1-3.3 ng·mL-1. The results show that this method is safe, accurate, reproducible and feasible, and could be used for the determination of anthraquinone derivatives in semen cassiae.In chpter 4:Dispersive liquid-liquid micro-extraction coupled with high performance liquid chromatographic method was established for the determination of anthraquinone derivatives in human urine after oral administration of San-Huang tablet. The kind and volume of dispersive and solvent extraction solvent, vortex and centrifuge time were investigated and optimized. Under the optimum conditions, the enrichment multiples are between 40-153. This method is safe, accurate, reproducible and feasible, and has been successfully used for the determination of anthraquinone derivatives in human urine. |
