The Effects Of BMP-2 On Bcl-2 And Bax Protein's Expression In HepG-2 Cell

OBJECTIVE:To investigate the effects of BMP-2 on protein Bcl-2 and Bax expression in the hepatic carcinoma HepG2 cell line in vitro.Its effective molecular mechanism to apoptosis of HepG2 cell was also observed.METHODS:0,1,10,100 ng/ml BMP-2 were added to HepG2 cell respectively.Western blot was used to observed the effects of BMP-2 on protein expression of Bcl-2 and Bax protein.All the experiment were repeated three times independently.RESULTS:1.The intensity of Bcl-2 protein band was(76.0±6.00)% in the HepG2 cell line before the BMP-2 treatment.But the intensity of Bcl-2 protein band was(63.0±4.36)%,(44.7±4.73)%,(39.7±3.51)%in the 1ng/ml,10ng/ml,100ng/ml BMP-2 intervention groups respectively. There was significant difference in the intensity of Bcl-2 protein band between BMP-2 intervention groups and control group(P＜0.05).There was also significant difference in the intensity of Bcl-2 protein band between 10ng/ml or 100ng/ml groups and control group(P＜0.01).2.The intensity of Bax protein band was(37.7±2.08)%in the HepG2 cell line before the BMP-2 treatment.But the intensity of Bax protein band was (46.3±4.73)%,(65.0±4.58)%,(84.7±7.23)%in the 1ng/ml,10ng/ml, 100ng/ml BMP-2 intervention groups respectively.There was significant difference in the intensity of Bax protein band between BMP-2 intervention groups and control group(P＜0.05).There was also significant difference in the intensity of Bax protein band between 10ng/ml or 100ng/ml groups and control group(P＜0.01).3.The rate of Bax/Bcl-2 was(49.5±2.59)%in the HepG2 cell line before the BMP-2 treatment.But the rate of Bax/Bcl-2 was(75.3±4.53)%,(145±5.08)%, (213.3±8.21)%in the 1ng/ml,10ng/ml,100ng/ml BMP-2 intervention groups respectively.There was significant difference in the intensity of Bax protein band between BMP-2 intervention groups and control group(P＜0.05).There was also significant difference in the intensity of Bax protein band between 10ng/ml or 100ng/ml groups and control group(P＜0.01).CONCLUSIONS:After intervention with the different conentration of BMP-2,the level of Bax in HepG2 was elevated obviously,while Bcl-2 expression was reduced obviously;and the rate of Bax/Bcl-2 was increased obvIous-ly.It indicates that BMP-2 promotes HepG2 cells apoptosis by altering the protein expression of Bcl-2 and Bax protein.
