Objective:To investigate the correlation of mast cells infiltrating in IgA nephropathy tubulointerstitial and to examine the relationship between mast cells and alphα-smooth-muscle-actin(α-SMA),proteaseactivated receptor-2(PAR-2)as well as sterm cell factor(SCF)expression. In addition,to investigate the probable mechamnism of mast cells infiltrating in IgA nephropathy tubulointerstitial damage.Methods:Renal biopsy specimens from 35 patients with a clinicopathological diagnosis of IgA nephropathy and normal renal tissue sections from 5 nephrectomized patients for nephroma were collected. Change of pathomorphological types was according to Lee SM and Katafuchi semiquantitative scoring system.The expression of tryptase, alph-smooth muscle actin(α-SMA),protease-activated receptor-2 (PAR-2)as well as sterm cell factor(SCF)were detected with immunohistochemistry.Possible correlation among the expression level of tryptase,α-SMA,PAR-2,SCF and histopathological score were evaluated.All data has been analyzed by SPSS 13.0.Results:1.The percentages of mast cells and the expressions ofα-SMA, PAR-2,SCF were increased with the aggravation of tub-uliointerstitial damage and the elevation of Lee grading in IgA nephropathy tubulointerstitial.2.The positive correlation is present positively between the percentages of MC①,the expressions level ofα-SMA②,PAR-2③, as well as SCF④(①/②r=0.887,①/③r=0.844,①/④=0.853,②/③r=0. 874,②/④r=0.887,③/④r=0.893,P<0.01).Also,there was a significant positive correlations between the score of renal tubule histopathological and MC,the expression level ofα-SMA,PAR-2,SCF.3.The positive correlation is present between the percentages of MC and serum creatinine(r=0.738,P<0.05).However,no correlation was seen between the percentages of MC and 24-hour urine protein. Conclusion:1.The positive correlation is present between the percentages of MC and tubulointerstitial damage in IgA nephropathy,sugesting that mast cells were probaly invovled in development of the tubulointerstitial damage in IgA nephropathy.2.Mast cell-derived tryptase,α-SMA,PAR-2,SCF were probaly invovled in development of the tubulointerstitial damage in IgA nephropathy.
