ObjectiveEvaluated the blood compatibility of several sorbents,To make some helps for the development of a new clinical effective hemperfusion sorbent to treat the endotoxemia.MethodsThere were assigned into five groups according to different adsorbents: group I:synthesis macroporous adsorptive resin of single use hemoperfutor from LIVZON corporation,groupâ…¡:ethyl cellulose with AMIKACINI ligands and coating with albumin,groupâ…¢:ethyl cellulose with PME ligands and coating with albumin,groupâ…£:ethyl cellulose with glutaraldehyde arms and coating with albumin,groupâ…¤:activated charcoal coating with albumin. Conducting four group materials including groupâ…¡,groupâ…¢,groupâ…£,groupâ…¤as samples and groupâ… as comparing,five materials were all tested with kinetic clotting time,APTT,platelet consumption rate,leucocyte consumption rate,erythrocyte consumption rate,C3 consumption rate,hemolysis rate.Then we evaluate the blood comparibility through the analysis of the above test results. Because the single and isolate parameter can't describe biomaterial's blood compatibility,we made use of the analytic hierarchy process to evaluate it systematically.Results 1.Through five materials were all tested with kinetic clotting time and APTT,it may conclude that the system of blood coagulation was partially activated according to biomaterials contacted with blood,sequentially,which induced in shortening clotting time.The biomaterials with good blood compatibility slightly affected the system of blood coagulation.In contrast to the groupâ… , The level of anticoagulation of groupâ…¡,groupâ…¢,groupâ…£is same which is between 40 and 50 minutes,and there is no statistical significance(P>0.05).While the anticoagulation time of groupâ…¤is between 20 and 30 minutes, groupâ…¤has lower anticoagulation comparing with the control and the difference of APTT is significant(P<0.05).2.The result of hemolysis test showed that the hemolysis rate of groupâ…¤was 8.8%which above the level of biomaterial(<5%),while the hemolysis rates of groupâ… ,groupâ…¡,groupâ…¢,groupâ…£respectively were 1.67%,1.24%,2.26% and2.67%which were all below 5%and they coincided with standard of biomaterials' hemolysis rate.The hemolysis rate by turns is groupⅡ<groupâ… ï¼œgroupⅢ<groupⅣ<groupâ…¤.3.The result of hemocyte consumption rate showed:there are no significant differences in the platelet consumption rate,leucocyte consumption rate and erythrocyte consumption rate of groupâ…¡,groupâ…¢and groupâ…£comparing with groupâ… (P>0.05),while groupâ…¤had higher hemocyte consumption rate(P<0.05). The platelet consumption rate by turns is groupⅡ<groupâ… ï¼œgroupⅢ<groupⅣ<groupâ…¤.The leucocyte consumption rate by tums is groupâ… ï¼œgroupâ…¢ <groupⅡ<groupⅣ<groupâ…¤.The erythrocyte consumption rate by turns is groupâ… ï¼œgroupⅡ<groupⅢ<groupⅣ<groupâ…¤.4.The result of C3 consumption rate showed:In contrast to the control,groupâ…¡, groupâ…¢and groupâ…£had low C3 consumption rate(P>0.05),however,the C3 consumption rate of groupâ…¤was higher(P<0.05).The C3 consumption rate by turns is groupâ… ï¼œgroupⅡ<groupⅢ<groupⅣ<groupâ…¤.5.The analytic hierarchy process used to evaluate biomaterial's blood compatibility of five materials showed:the synthetic scored index of five groups were respectively 41.7,35.4,30.8,30.2,12.4.The blood compatibility by turns is groupâ… ï¼žgroupⅡ>groupⅢ>groupⅣ>groupâ…¤.ConclusionsTo four sample materials,the blood compatibility of groupâ…¡is the best,then the sequence is groupâ…¢,groupâ…£.Compared with the groupâ… ,several single and isolate parameters of the three groups were insignificantly,while groupâ…¤had the lowest blood compatibility so as not to accord with the standard of adsorbent using in clinic.
