Objectives: The study aims to research the membrane proteins on liver sinusoidal endothelial cells both from normal liver and regenerating liver of rats (72h post partial hepatectomy) with subcellular proteomic methods so that we can further explore the relations between various hepatectomized size and liver regeneration.Methods: After having established the rat partial hepatectomy model in PHx group with Sham operation group (6 rats each group), we collected membrane samples at 72h after operation according to the technique of cationic colloidal silica membrane perturbation. Some samples were prepared for transmission electron microscopy. The profiles of membrane proteins expressed on liver sinusoidal endothelial cells both from normal livers and regenerating livers were analyzed with proteomic methods (separation by IDE SDS-PAGE and identification by MS/MS) and the proteins we had identified were inquired by bioinformatics methods.Results: We established the partial hepatectomy model successfully, all rats survived. Transmission electron microscopy results showed that by the technique of membrane perturbation only specified membranes of endothelial cell had been binding with cationic colloidal silica and the silica had being sticking to the sinusoidal endothelial cell membrane densely. We selected 24 straps from Sham group and PHx group altogether and identified 9,8 proteins from each group respectively.Conclusions: After having established the partial hepatectomy model in rats followed by subcellular proteomic research, we not only confirmed differential expression of membrane protein of sinusoidal endothelial cells at 72h post-hepatectomy, but also identified differential proteins both from normal liver and of regenerating liver. And we established the reference data of membrane proteins of sinusoidal endothelial cells, which had provided foundation for further study concerning various hepatectomized size and liver regeneration.
