Studies On The Chemical Constituents And Quality Control Of Sophora Flavescens Ait

AIMThis study was carried out to investigate the active chemicals in medicinal substance Kushen.,the dried roots of Sophora flavescens Ait,and elucidate the foundation of its pharmaceutical contribution,to provide the experimental basement of an overall quality control and global exploitation in medicinal substance Kushen.METHODS1.Extractions,polyamide and silca-gel column chromatography,and recrystallization were used in the processes of separation and purification.2.Physico-chemical properties and spectroscopic properties such as UV, IR,NMR,and MS were used in the structure identification.3.A HPLC method was used in the determination alkaloids and flavonoids in S.favescens Ait.RESULTS1.Eight compounds were obtained from the 95%ethanol extracts of S. favescens Ait.2.Oxymatrine(Ⅰ),matrine(Ⅱ),β-sitosterol(Ⅲ),lupeol(Ⅳ), l-maackiain(Ⅴ),trifolirhizin(Ⅵ),quercetin(Ⅶ)and rutin(Ⅷ)were elucidated o the basis of physico-chemical properties and spectral evidences.3.A HPLC method was established for the simultaneous determination of alkaloids(matrine and oxymatrine)and flavonoids(trifolirhizin)in S.favescens Ait.CONCLUSIONS1.Eight compounds were obtained from S.faveseens Ait.,among which lupeol(Ⅳ),quercetin(Ⅶ)and rutin(Ⅷ)were isolated for the first time.2.A HPLC method was established for the simultaneous determination of alkaloids(matrine and oxymatrine)and flavonoids(trifolirhizin)in S.faveseens for the first time.This method is simple,rapid and reliable,and provide an advancement in quality control with experimental basement of medicinal substance Kushen.
