Objective: to determine the express of HO-2 and eNOS in the corpus cavernosum of castrated rats and discuss the relationship between HO-2,eNOS and erectile function of penis in castrated rats. Method: 40 SD rats of 10 weeks were divided into 4,8,12 weeks castrated group,Which were castrated by ectomizing bilateral testis and the skin was only excised in the control group. serum testosterone were detected. All rats underwent erectile function testing in vivo by measuring intracavernosal pressure and mean arterial blood pressure upon electrical stimulation of the major pelvic ganglion. penis were ectomized , HO-2 was Analyzed by Western blot , eNOS and HO-2 were detected by immunohistochemistry. Result: The serum testosterone and ICP/ MAP decreased after castrated(P<0.05). HO-2 exits in all group, The area of under the curve of 4 weeks castrated group(341.50±99.70) is less than that of 8 weeks castrated group (705.00±152.74) and control group(876±443.36)(P<0.05). There were no significant changes between 8 weeks castrated group and control group. We didn't find HO-2 in the corpus cavernosum of 12 weeks castrated group. eNOS express in the astrated groups are less than that of control group apparently. Conclusion: Androgens might mediate the erectile function through HO-2,eNOS in the corpus cavernosum of rats.
