ObjectiveTo investigate the effect of administration with heme oxygenase-1induceron the expression of heme oxygenase-1(HO-1) and nitric oxide synthase(NOS)and the level of oxide stress and erectile function in rats with diabetic erectiledysfunction.Methods44Sprague-Dawley male rats,8rats in normal control(NC) group were asage control,36rats with diabetes mellitus erectile dysfunction(DMED) wereassigned randomly into three groups, consisting of DMED control group,Hemin group and Hemin+Znpp group; the rats in NC group and DMED groupwere treated only with vehicle by intraperitoneal injection, Hemin group weretreated with Hemin and Hemin+Znpp group were treated with Hemin and Znppby intraperitoneal injection, after4weeks administration, the serum glucoseand weight of all rats were recorded; the distribution and content of HO-1、eNOS and iNOS in the corpus cavernosum were determined byimmunohistochemistry and western blot analysis; MDA content and SODactivity in corpus cavernosum were measured by thiobarbituric acid assay andxanthine oxidase assay respectively; erectile function of rat was accessed byapomorphine assay.ResultsHO-1, eNOS and iNOS mainly express in the cytoplasm of smoothmuscle cell of corpus cavernosum and vascular endothelium of all rats.Compare to the DMED group, the expression of HO-1in corpus cavernosumof rats increased in Hemin group and Hemin+Znpp group(all P<0.05), but the Hemin+Znpp group was less than Hemin group(P<0.05); compare to DMEDgroup, the expression of iNOS and the content of MDA and the serumglucose declined in Hemin group and Hemin+Znpp group(all P<0.05), but theexpression of eNOS and the activity of SOD and the weight increased inHemin group and Hemin+Znpp group(all P<0.05); the expression of iNOSand the content of MDA and the serum glucose in Hemin+Znpp group werehigher than those in Hemin group(all P<0.05), but the expression of eNOSand the activity of SOD and the weight in Hemin+Znpp group were less thanthose in Hemin group(all P<0.05); Compare to the DMED group, the erectilefunction of rats were restored partly in Hemin group and Hemin+Znpp group,the erectile times of rats in Hemin group were more than Hemin+Znppgroup(P<0.05), but still less than NC group(P<0.05).ConclusionsTreatment with HO-1inducer could restore erectile function of DMED rats,which may related that HO-1inducer could increase the expression of HO-1,regulate the expression of eNOS and iNOS, ameliorate the oxidation stress incorpus cavernosum and improve glucose metabolism, suggesting that HO-1inducer may be an effective drug choice for patient with DMED.
