A Study On The Human Interferon-α-2b To The Treatment Of Endometriosis

Objective:The purpurse of this study was to approach the mechanisms and effects of human interferon-α-2b(IFN-α-2b)on the treatment of endometriosis(EMs)by measuring serum level of the cancer antigen 125(CA125)and antiendometrial antibody(EMAb)and observing the effects of the treatment by combining the surgery with human IFN-α-2b on endometriosis.Methods:Forty-five cases who had the endometrial cyst of the ovary and accepted the conservative operation in the department of gynecology of the second affiliated hospital of Tianjin medical university were selected,whom were 23-45years and diagnosed by histopathologist after operation.They were randomly divided into three groups.Test group(operation+IFN-α-2b),15 cases,Control group A(operation+ diphereline),20 cases,Control group B(only operation),10 cases.The methods of administration were as follows:Test group was given human interferon-α-2b 100,000U once every other day by intramuscular injection from the seventh day of post-operation,fifteen times;Control group A was given diphereline 3.75mg once every four weeks by intramuscular injection from the first day of menstrual onset, four times;Control group B was not given any management.Each cases was draw venous blood 2ml×2 on the pre-operation,the first month,second month and third month of the post-operation,centrifuged and taken the supematant and preserved at the -70℃.The CA125 level was measured by microparticle enzyme immunochemiluminescent and the EMAb level was measured by enzyme linked immunosorbent assay(ELISA).The follow-up was carried out after operation to evaluate the clinical appearances and recorde the adverse effects of the human interferon-α-2b.Results:1.The changes of CA125 level:Compared with pre-operation,the CA125 level of the first to third month after operation obviously degraded in test group and control group A(all P＜0.05),the CA125 level of the first and third month after operation did not obviously degrad in the control group B(all P＞0.05);Compared the test group with control group A,the CA125 level of the preoperation,the first month,second month and third month of the post-operation all wasn't significantly deviation (all P＞0.05);Compared the test group with control group B,the CA125 level of pre-operation and the first month of post-operation all had no significant deviation(all P＞0.05),But the CA125 level of the test group in the third month after operation was lower than that of the control group B.2.The changes of EMAb level:Compared with pre-operation,the EMAb level of the first to third month of post-operation obviously degraded in the test group and control group A,B(all P＜0.01);Compared the test group with control group A,the EMAb level of the preoperation,the first month,second month and third month of the post-operation all had no significant deviation(all P＞0.05);Compared the test group with control group B,the EMAb level of the preoperation had no significant deviation,however,the EMAb level of the first month after operation of the test group was lower than the control group B.3. In the follow-up,we observed the adverse effects of the human interferon-α-2b.The adverse effects were low-grade fever,articular muscle soreness and gastrointestinal tract complaints and so on,these appearances completely disappearaed after drug withdrawal,it had less impact on the menstruation.The treatment of combining triptorelin also emerged propotional adverse effects,but they lasted longer.Conclusions:This study observed the effects of the treatment combining surgery with human IFN-α-2b on endometriosis by monitoring the levels of CA125 and EMAb from the first to third month after operation.We discovered that the CA125 and EMAb level of the post-operation descended persistently.The effects of treatment of human IFN-α-2b on endometriosis are similar with those of GnRH-a, both of them are all better than the pure sugery.In addition the adverse effects of the human IFN-α-2b persist shorter.It has less impact on the menstruation and can avoid menostasia,hectic fever decreased libido and so on that caused by GnRH-a.The price is cheaper.So the human IFN-α-2b has extensive prospect of clinical application.The serum levels of CA125 and EMAb are valuable of evaluating and monitoring therapeutic efficacy of treatment on endometriosis.But the machanisims of action and long-term effect of human IFN-α-2b still need explore further.
