Objective: Angiogenesis is the base of the tumor growth, invasion and metastasis. It plays an important role both in primary and metastatic tumors. Heparanase, an endo-β- glucuronidase, plays an accessory role in angiogenesis and tumor development by releasing HS-bound growth factors and promoting basement membrane degradation. Thrombospondin-1 is an important extracellular matrix protein that inhitits endothelial cell proliferation, migration, and angiogenesis. Our study detected the expression of HPA mRNA and TSP-1 mRNA in the primary tumor and the tissues set apart 5cm from tumor of breast cancer in order to investigate the interaction of angiogenesis and antiangiogenesis, and to provide assistance for prognosis and treatment of breast cancer.Methods: 31 cases of breast cancer consisting of 24 infiltrating ductal carcinoma, 6 infiltrating lobular carcinoma, 1 papillary adenocarcinoma, were selected from the Forth Hospital of Hebei Medical university during May to August in 2007. Reverse transcription polymerase chain raction (RT-PCR) was used to detect the expressions of HPA mRNA and TSP-1 mRNA in the primary tumor tissue and the tissue set apart 5cm from the tumor of 31 cases of breast cancer, Immunohistochem -istry (S-P method) was used to detect the expression of VEGF, c-erbB2, ER, PR and microvessels density of breast cancer tissues, All results were statistically analyzed.Results: 1 Positive rates of HPA and TSP-1 mRNA were all higher in the primary tumor tissues than tissues set apart 5 cm from tumor(P<0.001, P<0.05 respectively).2 According to the size of the primary tumor, patients were divided into three groups: group 1:≤2cm, group 2: >2cm and≤5cm, group 3: >5cm. The positive rate of HPA mRNA in group 2 was higher than that in group 1(P=0.004), there was no difference between other groups (P>0.05). Positve rate of TSP-1 mRNA in group 1 was higher than which in group 2 (P=0.033), there was no difference between other groups (P>0.05).3 According to the condition of axillary nodes metastasis, patients were divided into three groups: group 1: the axillary nodes metastasis was negative, group 2: 1~3, group 3:≤4. The positive rate of HPA mRNA in group 3 was obviously higher than which in group 1 (P=0.004), there was no difference between other groups (P>0.05).The expression of TSP-1 mRNA was no difference in all groups (P>0.05).4 According to theΜICC TNM staging value (2003), patients were divided into stage I group, stage II group and stage III group. The positive rate of HPA mRNA of the group stage II and stage III group were higher than stage I group (P>0.05), there was no difference between stage II group and stage III group (P>0.05).The expression of TSP-1 mRNA was no difference in all groups(P>0.05).5 According to the patho-typing standard of WHO (2003), patients were divided into the infiltrating ductal carcinoma group and the infiltrating lobular carcinoma group. There was no difference between the two groups(P>0.05).6 According to the histio-staging standard of WHO (2003), patients were divided into three groups, the histological grade I group, grade II group and grade III group. There was no difference among them neither HPA group nor TSP-1 group (P>0.05).7 According to the state of tumor embolus in vessel, patients were divided into two groups, group of tumor embolus negative in vessel, group of having embolus in vessel. The positive rate of HPA mRNA was obviously difference between two groups(P=0.005). There was no difference between two groups in positive rate of TSP-1 mRNA(P>0.05).8 According to the expression of HPA and TSP-1 mRNA, patients were divided into two groups respectivly. There were obviously differences of the values of microvessel density between the groups (t=-4.513, P=0.000; t=2.557, P=0.016).9 According to the intensity of the expression of VEGF, patients were divided into two groups: group 1: VEGF was negative or (+), group 2: VEGF was (++) or (+++). There was no difference in the positive rates of HPA and TSP-1 mRNA between the two groups(P>0.05).10 According to the the intensity of the expression of ER, PR and c-erbB2, patients were divided into two groups: the (-) group and the (+) group. There were no difference to the positive rates of HPA and TSP-1 mRNA between groups of ER, PR and c-erbB2(P>0.05).11 According to the state of catamenia, patients were divided into pro-menopausal group and menopausal group. There were no difference in positive rate of TSP-1 mRNA between two groups (P>0.05).12 There was no correction between the level of HPA and TSP-1 in breast cancer tissues(r=-0.042,P=0.824)Conclusions: 1 The expression of HPA and TSP-1 in breast cancer is significantly higher than those in normal tissue set apart 5cm from tumor.2 The microvessel density in the tissue with HPAmRNA is higher than that with negative HPAmRNA. While it is opposite to TSP-1.3 The level of HPAmRNA in breast cancer is significantly increased with the enlargement of primary tumor, with the metastasis of axillary lymph nodes and with the advancement of the TNM stage.4 The level of HPA mRNA in breast cancer with tumor embolus in vessel, which may be promote the invasion and metastasis of tumor cells and predict the poor prognosis, is higher than that without tumor embolus in vessel.5 There is no correlation between TSP-1 mRNA expression and the metastasis of axillary lymph nodes and TNM stage. But TSP-1 mRNA expression is found to be correlation with the enlargement of primary tumor.6 The expression of HPA mRNA and TSP-1 mRNA are not found to be correlation with the expressions of ER, PR, c-erbB2 and VEGF in breast cancer, and tumor pathologic types, histological grade, and menopausal status.7 There is no obvious relationship between HPA and TSP-1 in breast cancer tissues...
