Multiple sclerosis (MS) is an autoimmune inflammatory demyelinating disease of the central nervous system. Although demyelination is the hallmark of this process, destruction of axons occurs as well. The diagnosis of MS is generally based on the combination of clinical, radiological and laboratory findings. Up to now, no single clinical feature or diagnostic test is sufficient to predict the subtype and progression of MS. However, the molecular mechanisms underlying the disease remain poorly understood. Cerebrospinal fluid (CSF) proteins may provide important information about the pathogenesis present in multiple sclerosis .In our study, using cerebrospinal fluid proteomics technol- ogy to find the biomarkers of MS and thus to study the pathogenesis of the disease.Methods1.Using the two-dimensional difference gel electrophores- is (2D-DIGE) technology for comparative analysis, we compared CSF samples from patients with MS of clinical definite MS or laboratory-supported definite MS (CDMS and LSDMS, n=5) and from patients with neuromyelitis optica (NMO, n =5) with neurologically normal controls(n =5). 2.We used matrix-assisted laser desorption ionization-time of flight–time of flight-mass spectrometry (MALDI-TOF- TOF-MS) to determine the abnormal CSF proteins in MS patients.ResultsOur data showed that the levels of eight proteins and their isoforms in CSF were significantly altered in MS patients compared with controls. Glutathione peroxidase, Chain A human serum albumin mutant R218h, Chain A crystal structure of the Ga module, Immunoglobulin kappa light chain, Immunoglobulin heavy chain, Catalytic antibody and PRO2619 were considerably increased in the CSF of MS patients, whereas haptoglobin was just increased in CSF of NMO patients.ConclusionWe concluded that these proteins may be involved in the pathogenesis of MS,but the role of these proteins still remains to be elucidated in detail and further validation is needed, and established fundament for explored the disease-specific markers of MS.
