Objective: To imitate livers with various degree of fatty infiltration by method of test tube with CT scan density calibrated (TCTDC),making them contrasted to the gray scale of these livers(GSL) by DFYПultrasound imaging analysis software(called for short below).And analyze the relation among GSL,CT scan density(CTD),CTD ratio between liver and spleen(CRLS)and hepatic fat content (HFC). As well, the relationship among GSL,body mass index(BMI),waist-to-hip ratio(WHR),fasting blood glucose(FBG) and blood fat index was analyzed. Therefore, the clinical value of DFY on quantitative diagnosis of fatty liver disease was studied.Methods: Livers with different fat content were imitated by TCTDC, measuring their CTD, and a correlation curve linked CT scan density (CTD) and the increasing percentage of fat content was derived. And through the curve, the corresponding CTD is found when HFC was 5% (called CTD5 for short ). 30 people with suffused fatty liver and CTD lower than CTD5 were selected, and their CTD ratio between liver and spleen(CRLS) and clinic index(BMI,WHR,FBG and blood fat index). Then comparing the standard curve, the corresponding HFC were found. Gray scales of these livers were measured by DFY, taking the maximum,minimum and mean value. Meanwhile, liver tissues of five patient were taken as Oil Red O method for lipofuchsin, calculating the fat content by volume percentage density(VP). The relationship among GSL,CTD,CRLS,homologous HFC and VP were analyzed, and the clinical value of DFY on quantitative diagnosis of fatty liver disease was investigated. .Results: (1) Through the method of TCTDC, the model's CTD had a negative linear correlation to the corresponding HFC (p<0.001).The CTD was 55.13Hu while HFC was 5%, and it was 44.44Hu while HFC was 10%. (2) The GSL measured by DFY were negative linear to CTD of the livers (P<0.001), and also negative linear to CRLS (P<0.05), while they were positive linear to HFC obviously (P<0.001). And CRLS were negative linear to HFC (p<0.001).(3) The GSL was 46.78 when HFC was 5%, and it was 85.24 when HFC 10%, and was 62.16 when 7%. GSL had an evident positive linear correlation to the homologous HFC(P<0.001), while HFC was super than 7%;however,they had no correlation found wile HFC was lower than 7% (P=0.859).And the variation argument of GSL was more obvious than that of CTD and CRLS. (4) DFY was better than simple ultrasound image on diagnosis of fatty liver disease(X=1,P<0.001).(5)Ultrasound attenuation of the livers had an conspicuous relation to HFC(P<0.001).(6)CTD was effected by BMI and WHR in some degree, while there was no effect on GSL.(7)The correlation coefficient of volume density percentage(VP) from pathological section analysis and HFC was 0.965(P<0.01), and they had an obvious correlation. As well, VP had an evident relationship to GSL, and the correlation coefficient was 0.964(P<0.01).Conclusion: (1) The GSL measured by DFY could reflex the fat content of liver finely. (2)There was an obvious relation on GSL and HFC while HFC>7%(GSL=62.16), So the critical point of diagnosis on fatty liver disease by DFY was when HFC being 7%(GSL=62.16). And while HFC was 7%-9%, the variation extent of GSL was more conspicuous than that of CTD and CRLS. Therefore, it was supposed that while HFC was 7%-9%, GSL may be better than CT on the quantitative diagnosis of development for fatty liver disease.(3)UA measured by DFY had an obvious positive relation to HFC, and it also could be used on imitate the fat content of liver.(4)GSL had no connection with BMI,WHR,BG and blood fat index, and was little influenced by them. (5)DFY maybe have an important pragmatic clinical value on diagnosis of pristine fatty liver disease and the follow-up of it.
