Objective:To observe the effect of Xinbitie transdermal patch acupoint application on anti-myocardial ischemia and investigate it's neural mechanisms on rabbit models.Methods:1.experiment partⅠ,40 rabbits was divided into 5 groups randomly, applied pituitrin or isoproterenol respectively causing the acute or subacute myocardial ischemia models.Light microscope,electron microscope, guide polysiological recorder,biochemistry,in situ end-labeling, immunohistochemistry and so on methods and in the cellular molecular level to discuss the combination mechanism of rabbits' myocardial damage on the process of occurrence,developments,and changes,from which get to know the effect and mechanism of Xinbitie transdermal patch acupoint application for anti-myocardial ischemia comparatively and deeply.2.experimentⅡ,24 rabbits divided into 3 groups randomly.To observe the neurophysiologic mechanism of Xinbitie transdermal patch on the acupoints for anti-myocardial ischemia under the condition of destruction on the nucleus of hypothalamus models,the pituitrin was used to prepare the acute myocardial ischemia model.Result:1.Compared with the other groups Xinbitie transdermal patch group could shorten the recovery time of electrocardiogram and had significant difference(p<0.05).2.Xinbitie transdermal patch group could reduce MDA and elevate SOD; the difference between Xinbitie transdermal patch group and nitroglycerin group was obvious(p<0.05).3.Caspase-3 gene expression of Xinbitie transdermal patch group was lower than the other groups(p<0.05).4.The excursion of ST potential ECG in Xinbitie transdermal patch group and impaired PVN group were lower than model group(p<0.05);the difference between Xinbitie transdermal patch group and impaired PVN group was obvious.(p<0.05)Conclusion:1.Xinbitie transdermal patch on the acupoints can improve the electrocardiogram appearance;diminish the damage of oxygen free radical, strengthen the ability of the rabbit on anti-oxidation;restrain and block the excess apoptosis of myocardial cell.2.PVN maybe one of central nerve nuclear mission which involve the process of Xinbitie transdermal patch acupoint application on anti-myocardial ischemia.
