Objective: To investigate the relations between salvia miltiorrhiza and HO-1when Rat ex vivo liver preserved ,further clear about the biological mechanism of salvia miltiorrhiza protective function to the liver of the rat,and propose theory basis of the salvia miltiorrhiza protective function to the liver in Ischemia/reperfusion injury.Method: 1. HO-1mRNA expression detected by Real-Time Reverse Transcription Polymerase Chain Reaction(RT-PCR) after the liver preserved in time compartments of each group;2. HO-1 protein expression level detected by Immunohistochemistry after the liver preserved in time compartments of each group;3. Observed morphous of hepatocyte and hepatic sinusoid endothelial cell by light microscope after the liver preserved in different time of each group;4. Evaluated the relation between HO-ImRNA and protein and liver injury by correlation analysis. Results:1. Rat ex vivo liver preserved mode had been succeefully established ;2.Result of RT-PCRdisplayed that The salvia miltiorrhiza causes HO-ImRNA expression up-regulation and microamount expression in group after ZnPP pretreatment; 3 Result of Immunohistochemistry displayed that The salvia miltiorrhiza causes HO-I protein expression up-regulation and microamount expression in group after ZnPP pretreatment; 4. Pathological slices may prove liver cell and hepatic sinusoid endothelial cell the most obvious injury can be seen in group after ZnPP and smallest injury in salvia miltiorrhiza group.5. It offer inverse correlation between HO-ImRNA expression and liver injury, positive correlation between gray scale of HO-1 protein and liver injury in same time of different group.Conclusion: The salvia miltiorrhiza may induce HO-1 HO-1mRNA and protein over expression, the relations of the over expression and the liver injury may be one of the machines of the salvia miltiorrhiza preserving ex vivo liver.
