Objective:To observe the effects of electroacupuncture on neuroethology and the expression of Caspase3 in ischemic penumbra in the early stage of cerebral ischemia in rats.It also provides academic foundation for clinical treatment and early rehabilitation.Method 100 Wistar rats were randomly divided into shamoperated group,operated group and electroacupuncture group.An experimental middle cerebral artery occlusion model established in operated and electroacupuncture groups rats,sham operation was carried out in the rats of shamoperated group without blocking medium sized artery of cerebrum,after operation,the rats of electroacupuncture group were treated with electroacupuncture.Immunocytochemistry was performed to detect the expression of Caspase3 in 24h,48h and 72h after cerebral ischemia in ischemic penumbra respectively,the behavioral neurological assessment of the rats were also performed.Result:1,The rats of the model groups and electroacupuncture groups show neurologic impairment in different degree after the model was made.The behavior of the rats of the shamoperated group is normal.2,Nerve function score show that there is significant differences between electroacupuncture groups and model groups in the same phase(P<0.05,P<0.01).There are significant differences between electroacupuncture-72h group and electroacupuncture-24 group (P<0.01).These show electroacupuncture can improve cerebral ischemia rats' nerve function and exist additive effect.3,the results show that:there was almost no positive expression in shamoperated groups.Many caspase3-positive cells were found in ischemic penumbra of the model groups and the electroacupuncture groups while the peak in 24 hours after cerebral ischemia.Significant increase of caspase3 expression were detected in rats of model groups and electroacupuncture groups at various time points after cerebral ischemia when compared with shamoperated groups (P<0.01).It was also found that the number of caspase3-positive cells of model groups were significantly more than those of electroacupuncture groups on 24h and 48h after cerebral ischemia(P<0.05). while there is no significant difference between model groups and electroacupuncture groups on 72h after cerebral ischemia.Conclusion:1,the model of MCAO was made successfully.2,electroacupuncture can improve the nerves function in the early stage,one of the mechanism is electroacupuncture can inhabit brain nerve cell apoptosis efficiently.3,The increase of caspase3-positive cells in ischemic penumbra was time-dependent,electroacupuncture can significantly decrease the expression of Caspase3.
