Objective(1)To investigate the effects of electroacupuncture(EA)on blood lipid,behavior,cerebral infarct volume,brain tissue morphology and expression of TrkA in hippocampal dentate gyrus and ischemic penumbra and expression of TrkA and ?-NGF in the cerebral hemisphere in Hyperlipidemia-Middle Cerebral Artery thrombus(hMCAT)rats.(2)To investigate the effects of EA on behavior,cerebral infarct volume and expression of TrkA and p-NGF in the cerebral hemisphere in Middle Cerebral Artery thrombus(MCAT)rats.MethodsExperiment 1:102 SD Rats were randomly divided into normal group(n=6),hMCAT group,sham operation group,EA 1 group and EA 2 group(n=24 in each group).Except for the normal group,the rats in other groups were fed with high-fat diet for 6 weeks.EA was applied at bilateral "Fenglong"(ST 40)on the rats in EA 1 group(2/100 Hz,20 min/day)from 43 to 49 day.At the 50th day,the MCAT were induced by 50%FeCl3(w/v)in model and EA groups.EA was applied at"Baihui"(GV 20)and bilateral "Fenglong"(ST 40)(2/100 Hz,20 min/day)on rats in EA groups immediately after MCAT,and applied once a day from 50 to 63 day.The levels of total cholesterol(TC),triglyceride(TG),high density lipoprotein(HDL)and low density lipoprotein(LDL)levels were detected 1 d,7 d and 14 d after operaton.Neurological deficit was assessed by modified Bederson's scoring and TTC staining was used to observe the cerebral infarction volume.Brain tissue morphology was detected with hematoxylin-eosin staining.Expression of TrkA protein in the dentate gyrus and ischemic penumbra detected by immunohistochemistry.The expression of TrkA and(?-NGF protein in the cerebral hemisphere was detected by Western blotting.Experiment 2:72 SD Rats were randomly divided into model group,sham operation group,EA 1 group and EA 2 group(n=18 in each group).All groups were fed with,ordinary fodder.EA was applied at bilateral "Fenglong"(ST 40)on the rats in EA 1 group(2/100 Hz,20 min/day)for 7 days.At the 8th day,the MCAT were induced by 50%FeCl3(w/v)in model and EA groups.EA was applied at "Baihui"(GV 20)and bilateral "Fenglong"(ST 40)(2/100 Hz,20 min/day)on rats in EA groups immediately after MCAT,and applied once a day from 8 to 14 day.Neurological deficit was assessed by modified Bederson's scoring and TTC staining was used to observe the cerebral infarction volume.The expression of TrkA and(3-NGF protein in the cerebral hemisphere was detected by Western blotting.ResultsExperiment 1:(1)The levels of TC,TG and LDL increased and levels of HDL decreased in the hMCAT group after operation(P<0.01,P<0.05).Compared with hMCAT group,TG in EA groups decreased at 7 days after operation(P<0.05),and LDL in EA 1 group decreased at 14 day after operation(P<0.05).The reduction extent in EA 1 group is bigger than that of EA 2 group.(2)Compared with hMCAT group,NDS in EA 1 group decreased at 1 day after MCAT(P<0.05).The infract volume of left brain in EA groups reduced(P<0.01).It was smaller in EA 1 group than EA 2 group(P<0.05).(3)In hMCAT group,large numbers of inflammatory cells aggregated and infiltrated in the ischemic penumbra.The nerve cells were swollen,severe nuclear damage indicated by pyknosis,neurons disappeared,and the nerve cells were loose.Compared with the hMCAT group,there were more surviving nerve cells,less irregular nucleuses in EA groups.The nerve cells were slightly swollen,the extracellular space was smaller,and the inflammatory infiltration was lighter.The edema of cells and interstitium was lightern The degree of brain injury in EA groups was lighter than that of the hMCAT group.It was lighter in EA 1 group than EA 2 group.(4)The number of TrkA-positive cells in the dentate gyrus and ischemic penumbra:At 1 day,7 day and 14 day after operation,the number of TrkA-positive cells was significantly increased in other groups compared with the sham-operation group(P<0.01).At day 1,compared with the model group,the number of TrkA-positive cells in the EA groups increased significantly(P<0.05,P<0.01).Compared with EA 2 group,the number of TrkA-positive cells rised significantly in EA 1 group(P<0.05).At 14 day,compared with the model group,the number of TrkA in the EA 1 group increased significantly(P<0.05).(5)The levels of TrkA and ?-NGF among groups had no significant difference after operation(P>0.05).The expression levels of of TrkA and ?-NGF had an increasing tendency at 1 day after operation and decreased to normal at 7 day after operation.They had an increasing tendency from 7 day to 14 day after operation in EA 1 group.They both had a higher expression in EA 1 group than EA 2 group.Experiment 2:(1)Compared with the MCAT group,NDS in EA 1 group decreased at lday after MCAT(P<0.05).The infarct volume of EA 1 group reduced(P<0.01).It was smaller in EA 1 group than EA 2 group(P<0.05).(2)The levels of TrkA and ?-NGF among groups had no significant difference after operation(>0.05).At 1 day after operation,compared with the sham-operated group,the expression level of TrkA in the MCAT group had an increasing tendency and the expression level of ?-NGF had an increasing tendency in the EA group 1 compared with the MCAT group.At 7 days after operation,the expression levels of TrkA and ?-NGF increased in the MCAT group compared with the sham group.Compared with the MCAT group,the expression levels of TrkA and P-NGF had an increasing tendency in the EA 1 group.Conclusion(1)Acupuncture intervention in the hyperlipidemia stage can improve the neurological function,reduce the volume of cerebral infarction,and improve the pathological morphological changes of brain tissue by regulating the blood lipid level,which is superior to the post-ischemic treatment.The possible mechanism is that the electro-acupunctre on "Baihui" and "Fenglong"can promote the expression of TrkA and ?-NGF.(2)Early electroacupuncture intervention can improve the neurological function and decrease the cerebral infarction volume in rats with cerebral ischemia.Early electroacupuncture intervention may up-regulate TrkA and ?-NGF in the cerebral hemisphere of cerebral ischemia model rats.(3)The intervention of electroacupuncture before cerebral ischemia has better effect.Acupuncture pretreatment is more effective than acupuncture after brain ischemia."Huatankaiqiao" acupoint compatibility method is much important in clinic.
