Objective:1. To study the expression of heparanase in T24 cells both in mRNA level and in protein level.2. To study the inhibit effect of the special siRNAs on heparanase expression.3. To study the effect on cell proliferation after siRNA downregulating heparanase (HPA) gene on bladder cancer cell line-T24.Methods:Chemically synthesized double stranded RNA (dsRNA) targeting heparanase gene was transfected into human bladder cancer cell lines T24. The proliferation of T24 cells was detected by MTT assay.( MTT methods was used to detect the proliferation inhibitory rate (IR)). Expression of heparanase mRNA was detected by reverse transcription polymerase chain reaction (RT-PCR) and Western-blot.Results:It was found heparanase had a high expression in human bladder cancer cell line-T24 both at mRNA and protein levels. MTT showed that it could suppress the growth of T24 cells to (28.55±3.66)%, (27.20±3.25)% and (37.29±4.82)% in three different groups. RT-PCR analyses found that there was a significant decrease in heparanase mRNA level in some experimental groups compared with those in control groups. It was showed that the expression of Hpa in T24 cells was down-regulated by siRNAs, both at mRNA and protein levels.Conclusions:The expression of heparanase is high in human bladder cancer cell line-T24. The proliferation of T24 cells and mRNA level of heparanase gene in T24 cells decrease after RNA interference (RNAi) on heparanase gene of T24 cells. Heparanase may be a key enzyme in invasion and metastasis of bladder carcinoma and may be a new target in cancer therapy.
