The fruit bodies of Auricularia auricular are one of the most important edible fungi in our country. Auricularia auricular is rich of polysaccharides that has multiple pharmacological activities. Recently, Auricularia auricular polysaccharide (AAP) is being paied great attention by scientists because of its significant health protection functions. In this dissertation, the author conducted a series of study on Auricularia auricula polysaccharides, which were isolated and purified from Auricularia auricula, and their physicochemical characteristics and chemical structure were determined with instruments analysis technique and chemicalmodification technique of biomacromolecule, and their hypoglycemic activity was also investigated with medical analysis and biological active analysis techniques.The ultrasonic process conditions on extracting water solube polysaccharides of Auricularia auricula were as follows: water extracting time is 123.52 minutes, ultrasonic output power is 561.11w, ultrasonic action time is 25.76 minutes, solvent: matter is 111.44:1(mL/g). Auricularia auricula was extracted by water with ultrasonic assistance and ethanol precipitated to produce crude Auricularia auricula water-soluble polysaccharides. Then it was refined in turn through deproteinization using Sevag method, decolorization by hydrogen peroxide and dialysis. The content of polysaccharides is 41.70%. AAP3-1 was absorbed by ion exchange chromatography of DEAE Sephadex A-25 and gel filtration chromatography of Sephadex G-200. The purity of refined Auricularia auricula water-soluble polysaccharides was checked by gel filtration chromatography of Sephadex G-200 and gel permeation chromatography (GPC). The MW of AAP3-1 which were determined with gel permeation chromatography are 1.19×104. The content of polysaccharides in AAP3-1 is 81.05%. Infrared ray spectrum of AAP3-1 also indicates that there are typical characteristic absorption peaks of polysaccharides. The preparation of sulfated Auricularia auricular polysaccharides(SAAP) has been preliminary investigated. The hyperglycemic function of AAP3-1 and SAAP was been compared. We can draw a conclusion that AAP3-1 and SAAP can reduce the serum glucose level of nomal mice. At the same time AAP3-1 and SAAP increase the liver glycogen. AAP3-1 and SAAP could cure diabetes mice induced by alloxan. After intaking AAP3-1 and SAAP for 6d continuously, AAP3-1 and SAAP can reduce the serum glucose level of hyperglycemic mice and increase the liver glycogen. SAAP has strong lowered serum glucose level than the AAP3-1. With lowered level of the serum glucose, AAP3-1 and SAAP can accelerate the glucose transform to liver glycogen. AAP3-1 could reduce the oral glucose tolerance test curve. Increasing the synthesis of liver glycogen and accelerating the anabolism of sugar may be one path of lowering serum glucose level.
