| ObjectiveAs a natural active polysaccharide,AAP has been proven to have a wide range of physiological regulation functions,including hypolipidemic effects;there are few reports on the preventive effect of AAP on hyperlipidemia and its mechanism of hypolipidemic action.In this thesis,we established a mouse model of hyperlipidemia and a rat model of prophylactic administration to investigate the prevention and treatment of polysaccharides from AAP on hypolipidemic and its possible mechanism,and provide reference for the prevention and treatment of hyperlipidemia.MethodsA female model of hyperlipidemia was established by ingesting high-fat diet for 5 weeks in female Kunming mice.The mice were given different doses of AAP for 4 weeks.The body weight of the mice was observed and the blood lipids were measured.The hypolipidemic effect of polysaccharides;the expression of inflammatory factors in serum and liver tissues was detected by enzyme-linked immunosorbent assay,and the level of antioxidant index in liver was determined.SD female rats were fed freely with high-fat diet,and a certain dose of AAP was administered by intragastric administration for 4 weeks(prophylactic administration).Blood lipids were measured,lipid metabolism enzymes and liver pathological changes were detected,and black fungus polysaccharides were verified.Whether it has a preventive effect on hyperlipidemia in the body,and explore its possible mechanism of preventing hyperlipidemia.Results1.AAP acute toxicology study results showed that after a large dose of AAP in seven consecutive days,the mice did not die,their mental state and activity were normal,appetite increased,female dose group and male400 mg/kg.bw dose group The weight gain was significantly lower than that of the normal control group(P<0.05),but the levels of WBC,RBC,HGB and PTL in the blood cells of male mice were significantly increased(P<0.05).2.The results of AAP on hypolipidemic effect in hyperlipidemia mice showed that the model of hyperlipidemia was established,and the body weight and body weight growth rate of mice decreased significantly after AAP treatment(P<0.05).Serum TC The level of LDL-C was decreased(P<0.05),and AAP was able to inhibit the expression of inflammatory factors TNF-α and IL-6 in serum(P<0.05)and reduce the level of NO.3.The preventive effect of AAP on hyperlipidemia in rats with highfat diet showed that after prophylactic administration of AAP in high-fat diet,body weight loss was slowed down,TC and LDL-C were decreased(P<0.05),TG Falling,HDL-C rises.After prophylactic administration of AAP in high-fat diet rats,the activity of lipid metabolism enzyme LACT enzyme was enhanced(P<0.05),which promoted the rate of systemic cholesterol transport,accelerated the clear rate of cholesterol in the liver,and decreased HMG-CoA activity.(P<0.05),the rate of cholesterol synthesis in the liver was reduced,and the effect of lowering TC and increasing HDL-C to prevent hyperlipidemia was achieved.Histopathological observation of the liver of the rats showed that the liver cells containing lipid droplets in the liver cells of the AAP group lost their intrinsic morphology,the number of cells decreased,and the liver cells containing lipid droplets did not exceed 3/4.Conclusion:Animal experiments show that AAP has a certain hypolipidemic effect in vivo,and can improve the degree of hepatic cell lipidosis.The mechanism of lowering blood lipids may inhibit the secretion of inflammatory factors such as TNF-α,IL-6 and NO in serum.In addition,in animals,the mechanism by which AAP prevents hyperlipidemia may be related to AAP directly reducing HMG-CoA enzyme activity or enhancing LACT enzyme activity. |
