| This test is aimed at the facts that the copper abuse by the domestic feed producers and the breeding industry.In this test,we took the KM mouce as animal models and exposured to copper by intragastric administration to being on the 30d accumulative toxicity experiment and metabolized & eliminated experiment.By doing the systematic research on the effects of copper to the animals' growth performance,physiology function,copper residue in tissue and in blood,even the copper elimination in mouse's tissues and blood,to provide some theoretical evidence and reference for the non-polluted meat products and the by-products even for the food supervision department.108 KM mice were divided into 2 batches by randomicity.The first batch had 48 mice divided into 4 groups named as control group,â… ,â…¡andâ…¢.Each group contained 6 female mice and 6 male mice.It made the CuSO4·5H2O as the copper source,exposure to copper by intragastric administration.Which three groups were given dose concentration as 10mg/kg,20mg/kg,40mg/kg by intragastric administration in the 30d accumulative toxicity experiment.The control group was given equivalent normal saline.We measured the mouse's growth performance during the experiment and tested the differential leukocyte count(DLC) with the blood samples collected form the tail vein each 10 days.After 30d,all the mice were killed to do pathological observations of organs and tissues and calculate the viscera index,even to test the blood biochemical index and detect the copper residue in tissues(liver,kidney, muscle) and in blood. The second batch had 60 mice divided into 5 groups by randomicity.Each group contained 6 female mice and 6 male mice.One of the groups was control group and the other 4 test groups were exposure to copper at the dose of 20mg/kg every day by intragastric administration for 10d.The control group was given equivalent normal saline. According to the scheduled sampling plan,the mice were killed after 0.25h-16d to test the copper elimination in liver,kidney,muscle and blood.The results of the experiments show that the 10mg/kg copper level can result in the mouse's growth performance improving.The total weight gain of this group's mouse was 14.84g compared with the control group 11.20g and the weight gain rate was 66.10% (control group 49.78%).The total feed intake was 151.08g(control group 135.19g).The total F/G was 10.18(control group 12.07).The 20mg/kg and 40mg/kg copper level reduced the mouse's growth performance.The 10mg/kg,20mg/kg,40mg/kg copper level had significant effect on the lymphocyte and neutrophil which result in the decrease of lymphocyte proportion and the increase of neutrophil proportion.The serum AST,ALT and LDH increased as the copper level adding.The 10mg/kg and 20 mg/kg copper level can significant enhance AKP but the 40mg/kg resulted in AKP increase.The 10mg/kg,20mg/kg,40mg/kg copper level can damage the heart,liver,spleen and kidney in different degrees and as the dose increased,the damage degree risen.The liver and kidney were the target organs for the toxic injury of copper.The liver and kidney were the main organs for the copper residue.The copper dose has good correlation with liver and kidney copper contents.The test groups' liver and kidney copper contents were significant different(p<0.05) or very significant different(p<0.01) compared with the control group.In the accumulative toxicity experiment,the liver copper contents inâ…¡(20mg/kg),â…¢(40mg/kg) groups and the kidney copper contents inâ…¡(20mg/kg) group were over 10mg/kg,which were higher than the National Residue Limit(10mg/kg) in the meat industry.The copper contents in muscle and in blood changed relatively stable as the copper dose increased. The metabolic time of copper was short in blood,and after 4h,the contents reduced to normal level.The metabolic time of copper was relatively long in liver and kidney,which reduced to normal level need about 96h and 192h.Copper content was relatively stable in muscle.The gradation of copper contents to peak value in mouse's tissues and blood was: liver(0.5h)>blood(1h)>kidney(2h)>muscle.ADVICE:please don't blindly increase the copper dose to diet in Livestock and Poultry Raising and do not treat the copper as the omnipotence additive in the Livestock and Poultry Raising in order to avoid the food safety problems that caused by the copper residue. |
