Objective:To seek the central target that Baixiangdan capsule cure PMS with liver-qi invasion by researching the effect of the medicine on the expression levels and location of gamma aminobutyric acid receptor A(GABA_AR)β2 subunit in cortex, hippocampi,hypothalamus and marginal of PMS with liver-qi invasion rats.Methods: PMS model rats with liver-qi invasion were prepared by electric stimulating.The protein expression levels of GABA_ARβ2 subunit in the cortex,hippocampi,hypothalamus marginal were detected with western blotting.The location of the above receptor were detected by labeling and observed with fluorescencemicroscopy.Results:Compared with the normal group,the expression of GABA_ARβ2 subunit increased markedly in the cortex,hippocampi and marginal(P<0.01) and the hypothalamus(P<0.05) of model group.The expression level of GABA_ARβ2 subunit decreased markedly in the former three tissues of Baixiangdan capsule group comparing with the model group (P<0.05),and it decreased insignificance in the hypothalamus.Compared with the normal group,the location of GABA_ARβ2 subunit had little changes in the cortex, hippocampi of the model and medication groups,but it changed markedly in the hypothalamus and marginal.GABA_ARβ2 subunit in the cortex,hippocampi of all groups distributed primarily in the fringe,in model and medication groups it changed little. GABA_ARβ2 subunit distributed dispersedly inside hypothalamus and marginal of normal rats,but distributed only in the edge in the two tissues of model and medication groups.Conclusions:It might be one of the most important cause resulting in PMS with liver-qi invasion that GABA_ARβ2 subunit expression levels increase markedly in the cortex,hippocampi,hypothalamus and marginal.Baixiangdan capsule can enhance the level of the three tissues markedly,but haven't any remarkable effects in location of the receptors.GABA_ARβ2 subunit can be the action target that the medicine cure PMS with liver-qi invasion in the special kind of cortex,hippocampi,hypothalamus and marginal.
