Objectice:To investigate the effects of diesel exhaust particles in the air pollutants inhaled to immediate asthma reaction in the mice.Methods:Wistar mice were randomly divided into 6 groups.There were 10 mice of each group.Group A was control group which were injected three times during three weeks and continuously inhaled saline for 7 days.Group B was animal model group which were injected to Ovalbumin(OVA)100mg+AL(OH)3 0.5ml on days 1,8,15 and inhaled with 3% OVA+saline for 30 min for 7 days.Group C,D,E,F were sensitized to Ovalbumin(OVA) and challenged with 3%OVA during 4 weeks,thus,inhaled DEP200μg/ml+ saline for one week,two weeks,three weeks,four weeks, Evaluate RL of each mouse,observe the number of eosinophils cells in brouchialveolar lavage fluid(BALF),and the pathological changes of the murine lung tissue.The concentration of IgE,interferon-γ(IFN-γ) and interleukin-5(IL-5) of the lung tissue were detected by ELISA.Results:①There was no airways hyperresponsiveness in the control group.The airways hyperresponsiveness were significantly different between the control group and each other group in the different time spots of 30 min(F=160.646,F=148.901,F=146.644,F=162.204,F=160.935, F=156.186,P<0.01).Group B was significantly different with group E and F(P<0.01).RL increased with the time of DEP inhaled(r=0.948,P<0.01).②The eosinophils counts of group B in the brouchialveolar lavage fluid were significantly greater than the control group(F=23.240,P<0.01).The eosinophils counts of group B was significantly different with group E and F(F=23.240,P<0.01).Neutrophlis of The major inflmmatory cells were infiltrated after DEP inhaled for one week.The Nentrophlis counts of group B was significantly different with group E and F(F=36.239,p<0.01).③The concentration of IgE was significantly different between group A and B(F=2.490,P<0.01),The concentration of IgE of group B was no significantly difference with group C,D,E,F(P>0.05).④The lung pathology showed the epithelial shedding,infiltration of peribronchial inflammatory cells and fibrosis on the basement membrane gradually.⑤The expression of IL-5 in the lung tissue gradually Increased from group C beginning.The expression of IL-5 of group B was significantly different with group C and E(F=4.263,P<0.01).The expression of IFN-γwas no significantly difference during every group(F=1.193,P>0.05).Concusion:Inhaled DEP in the air pollutants may exacerbate immediate asthma reaction in the mice.
