Objective To investigate the effects of Diesel Exhaust Particles inhaled to late asthmatic reaction in the mice.Methods Wistar mice were randomly divided into 6 groups,There were 10 mice of each group.Group B was asthma animal model groups which was sensitized by peritoneal injection to 100mg Ovalbumin(OVA)+100mg AL(OH)3 in the 1,8,15 days. Group A was control group which challenged saline that replacing the OVA,the step is the same to the group B.Group C,D,E,F respectively challenged with OVA,the step is the same to the group B,then inhaled DEP200μg/ml+ saline for one week,two weeks,three weeks,four weeks.After a week inhaled 3%ova+saline 30min again. After ova were challenged,Methacholine(MCH) was taken for the bronchial provocation test(BPT) in all groups through 24 hours.Evaluate RL of each mouse; Observe the number of eosinophils Cells in brouchialveolar lavage fluid(BALF);The concentration of IgE,interferon-γ(IFN-γ) and Interleukin-5(IL-5) of lung tissue were detected by ELISA;Observe the pathological changes of lung tissue.Results①There was no airways hyperresponsiveness in the control group.The airways hyperresponsiveness were significantly different between the control group and each group(F=104.472,P<0.01) And increase gradually with the inhalation of the DEP(r=0.886,P<0.01).The airways hyperresponsiveness of group E and F was significantly different with group B(P<0.01).②The eosinophils count of group B and C,D,E,F in brouchiaiveolar lavage fluid were significantly greater than the control group(P<0.01).The inflammatory cells were infiltrated after DEP inhaled for one week.The eosinophils count of group E and F was significantly different with group B(P<0.01).③The concentration of IgE was significantly different between group B,C,D,E,F and A(F=54.611,P<0.01),And increase gradually with the inhalation of the DEP(P<0.01),The expression of IFN-γwas no significantly difference between the group C,D,E,Fand group B(P>0.05).④The IL-5 was significantly different between group B,C,D,E,F and A(F=23.667,P<0.01) And increase gradually with the inhalation of the DEP(P<0.01).The IL-5 was significantly different between group D,E,F and B(P<0.01),The was IL-5 density depending on the inhalation time of dep(r=0.583,P<0.01):The IFN-γwas significantly different between group B,C,D,E,F and A(F=33.229,P<0.01) And increase gradually with the inhalation of the DEP(P<0.01).The IFN-γwas significantly different between group E,Fand B (P<0.01),The was IFN-γdensity depending on the inhalation time of DEP(r=-0.550, P<0.01).⑤The lung pathology showed the epithelial shedding,infiltration of peribronchial inflammatory cells and fibrosis on the basement membrane gradually.Conclusion Inhaled DEP caused asthmatic inflammatory changes by TH 2 lymphocyte,And increase gradually with the inhalation of the DEP(P<0.01).
