Objective:Observe the effect of granula of ZhuangGuJianXi on nitric oxide and prostaglandin E2 content in the synovial fluid, histomorphology in articular cartilage and synovial, apoptosis of articular chondrocytes of osteoarthritis in knee of rabbits, meanwhile study on analgesia and anti-infammation and anti-swelling effects of granula of ZhuangGuJianXi, discuss the mechanism of granula of ZhuangGuJianXi prevent and treatment to osteoarthritis, provide a cheap and effective drug for clinic, lay a good foundation for declaration of new traditional Chinese drug.Methods:1. Analgesia of pharmacodynamics study on granula of ZhuangGuJianXi by using the model of ear swelling of mice was adopted to assay.2. Study on anti-infammation and anti-swelling effects of granula of ZhuangGuJianXi by using the pain model of mice was induced by acetic acid writhing.3. Replicating knee osteoarthritis by modified Hulth's modeling method , study on the effect of granula of ZhuangGuJianXi to histomorphology in articular cartilage and synovial, meanwhile determination of nitric oxide and prostaglandin E2 content in the synovial fluid. Apoptosis of adenocarcinoma cells was evaluated by the method of tunnel, study on the effect of granula of ZhuangGuJianXi to apoptosis of articular chondrocytes.Results:1. In respect of the writhing times and latent time, very significant contrasts are found between low dose group of Granula of ZhuangGuJianXi, middle dose group, high dose group, control group and the model group (P<0.01), significant contrasts are found between , middle dose group, high dose group and low dose group of Chinese traditional (P<0.05 ) . 2. In respect of the degree about ear swelling, very significant contrasts are found between low dose group of Granula of ZhuangGuJianXi, middle dose group, high dose group, control group and the model group (P<0.01), middle dose group, high dose group and low dose group of Chinese traditional (P<0.05) .3. Results of pathology suggested that: In normal group, the appearance of joint capsule is normal, kiyosuke, articular surface is smooth, milkiness, bright, no cracks and defects was found, synovial cell is regular arrangement, no edema in intercellular matrix was found, the arrangement of chondrocyte and distribution of matrix with laws, superficial cells is spindle, level arrangement, intermediate cell is round, scattering distribution, chondrocyte of radiation layer cylindrical distribution; In model group, joint capsule gets to thickening, articular surface erosion with local cartilage defect, the edge of cartilage destruction and steophyte hyperplasia, capillary hyperplasia under synovium, edema in intercellular matrix, lymphocyte and plasmocyte infiltration, interspersed hemorrhage were found, superficial layer of cartilage gets to thinning and goes rough, superficial cells disappeared, arrangement of chondrocyte and distribution of matrix is irregular, deep cell have phenomenon of hyperplasia, structure of cartilage gets to disorderring, tide mark is incomplete; In control group, articular surface is smooth with the interspersed depression, superficial layer of cartilage gets to thinning, but the facet is smooth, some have the small crack, superficial cells gets to disappearing, chondrocyte of rediatum layer gets to atrophicing and the levels of cell is disorde; In low dose group of Granula of ZhuangGuJianXi, articular surface is not smooth, femoral condyle is coarse with some osteophyte generation, epithelial cells have mild hyperplasia, edema in intercellular matrix, lymphocyte infiltration, superficial cells gets to disappearing, chondrocyte of radiation layer gets to analosisinga and the levels of cell is disorder; In middle dose group, articular surface have interspersed depression, no obvious erosive local cartilage defect, synovial cells arrangement monolayer, no hyperplasia of epithelial cells, no edema in intercellular matrix, little capillary hyperplasia, arrangement of chondrocyte is regular, karyopyknosis in chondrocyte were found and chondrocyte proliferat; The results of high dose group lie between the low dose group and middle dose group, the degeneration of cartilage were less than control group.4. In respect of the scores with Markin's scoring, very significant contrasts are found between low dose group of Chinese traditional, middle dose group, high dose group, control group and the model group (P<0.01) , significant contrasts are found between middle dose group and the control group.5. In respect of the contents about NO and PGE2, significant contrasts are found between low dose group of Chinese traditional, middle dose group, high dose group, control group and the model group(P<0.05, P<0.01), significant contrasts are found between middle dose group, high dose group and the control group.6. In respect of the apoptosis index about chondrocyte, very significant contrasts are found between low dose group of Chinese traditional, middle dose group, high dose group, control group and the model group(P<0.01, P<0.05),significant contrasts are found between low dose group of Chinese traditional, middle dose group, high dose group and control group (P<0.05 ) , significant contrasts are found between, middle dose group, high dose group and low dose group of Chinese traditional.Conclusions:Granula of ZhuangGuJianXi have significant inhibitory effect on acute inflammation and pain, it can prevent and cure osteoarthritis of the knee through improving cartilage degeneration, restraining inflammation of synovium ,reducing secretion of inflammatory mediator, decreasing apoptosis of articular chondrocytes, middle dose group, high dose group is the best.
