Objective:To explore the effect of Guanjiezhitong capsule and stem cell gel complex on the repair of rabbit articular cartilage defect.Methods:BMSCs were isolated,purificated and raised from bone marrow by density gradient centrifuge and adherence culture method.BMSCs at passage 3 were selected to composite with fibrin gel.In the experiment study,40 New Zealand white rabbits altogether 80 sides knee joints,were randomly divided into four groups: Group I implanted without any complex into the defects. Group II implanted with stem cell gel complex without cytokines.Group III and Group IV implanted with stem cell gel complex with cytokines TGF-βand IGF-I. The rabbits inGroup II and Group IV were gavaged Guanjiezhitong capsules daily. The rabbits were sacrificed at 4,8,12 weeks.Each specimen was evaluated by both morphology and histology.Results:BMSCs raise so fast that we can obtain enough cells to repair the defect of articular cartilage. At 12 weeks after operation, cartilage defects of group IV were completely filled with hyaline cartilage. Chondrocytes arranged regularly.The thickness of the repair tissues was basically the same as the normal cartilages.Conclusion : Guanjiezhitong capsule and stem cell gel complex can repair cartilage defects effectively.In vivo,Guanjiezhitong capsules can induce BMSCs into chondrocytes. It can promote cell-gel complex repair cartilage defects.
