Objective To discuss the effective of autoallergic cancellous bone as an antibiotic carrier and analyze the release pharmacal regularity of antibiotic-cancellous bone in vitro processed with the purpose of giving further information for its clinical application.Methods Collection several human femoral heads after hip arthroplasty under sterile conditions,the samples was morselized of 175 bone blocks about 1cm×1cm×0.5cm at size and the mean weight of bone was 1.0g.The bone blocks were randomly divided into 4 groups.Briefly all bone was impregnated with three antibiotics:Gentamicin Sulfate,Ciprofloxacin Levo,Cefotaxine Sadium in 20ml for 10 minutes.First,the concentration of antibiotics eluted from the antibiotic-impregnated bone was measured after 3rd,6th,12th,24th,48th hours in high performance chromatogram of liquid assay.Second the antibiotic-impregnated bone was cultured in correspoding Staphylococcus aureus,Pseudomonas aeruginosa,Bacillus coli for 24 hours,each group the inhibition bacterial growth zone was measured with sliding caliper and the effect was draw curves.Results 1.Medicine concentration measurement:the medicine concentration was in the highest level in third hour.And subsequently the concentration gradually decline corresponding with time prolong,display slowly degrade.The concentration diversity obviously in 24th hour.The concentration of Sulfate Gentamicin was measured to be higher than the minimum inhibitory concentration of the SE for 48 hours and the cancellous bone was of a good sustained release feature.2.Antibacterial experiment in vitro:the curves of three antibiotic antibacterial is similar,the inhibition bacterial growth zone diametre gradually decline accordingly with the time prolong.The zone diametre change obviously for 24 hour and can be measured at 48th.The study showed cancellous bone as an antibiotic carrier have a fine releasing property and could gratify local antibacterial after eptic operation.Conclusions Cancellous bone as antibiotic carrier was of a good sustained release feature in vitro,local anti-infection and osteogenesis.It might be an ideal antibiotic carrier material.
