Objective: To explore effects and mechanism of Traditional Chinese Medicine BuShen HuaYu JieDu(BSHYJD) recipe on spontaneous lung metastasis in mice Lewis lung cancer. So as to provide scientific experimental evidence for clinic applying BSHYJD recipe in lung cancer treatment and especially preventing metastasis of lung cancer in late stage.Methods: Lewis lung cancer cell was implanted into 48 C57BL/6J mice, so Lewis lung carcinoma model in mice model was established. After 24 hours later, mice were divided into 6 groups randomly(n = 8 each): Control group, DDP group, Middle dose of BSHYJD, High dose of BSHYJD, Middle dose of BSHYJD with DDP, High dose of BSHYJD with DDP , After the BSHYJD recipe was given continuously by intragastric administration for 21 days and the chemotherapy drug was given by intraperitoneal injection for 3 days, the mice were killed by decapitation. We weighed the lung carcinoma, the spleen and thymus, observed the lung cancer metastasis lesions with Anatomy microscope , and calculated the rate of inhibitory effect of lung carcinoma and lung metastasis. The experiment was made to observe effects of Traditional Chinese Medicine BuShen HuaYu JieDu(BSHYJD) recipe on spontaneous lung metastasis in mice Lewis lung cancer and immunologic function of Lewis lung cancer mice, Meanwhile, we measured mtp53 and nm23 gene expression of the C57 mice through Immunohistochemistry technique and real-time fluorescence quantitative Reverse Transcription -Polymerase Chain Reaction (RT-PCR) to explore the mechanism of Traditional Chinese Medicine BuShen HuaYu JieDu(BSHYJD) recipe on spontaneous lung metastasis in mice Lewis lung cancer.Results: It has showed out that BSHYJD recipe has remarkably suppressing effects of mice Lewis lung cancer and metastasis lesions, and it can significantly increase the inhibition rate of DDP to transplanted Lewis lung cancer and the lung cancer metastasis(compared with control group, P<0.05), meanwhile there has evidently enhancement of immunologic function for BSHYJD recipe in lung cancer treatment, especially for High dose of BSHYJD with DDP(H+X) (compared with control group, P<0.05). The BSHYJD recipe could increase the expression of mRNA of nm23 gene and decrease the expression of mRNA of mutant-type p53 gene(compared with control group, P<0.05), and the BSHYJD recipe with DDP group which compared with the DDP group and model group could have statistic difference(P<0.05).Conclusions: The BSHYJD recipe can inhibit obviously the growth and spontaneous metastasis of Lewis lung carcinoma in mice, and enhance effects of chemotherapy-therapy. The mechanism of this may be correlated that BSHYJD recipe could increase the expression of mRNA of nm23 gene and restrain the expression of mRNA of mutant-type p53 gene. The BSHYJD recipe can also enviously enhance the immunologic function of Lewis lung cancer mice.
