Objective:To investigate effects of the psoralen on cell proliferation of osteoblast,ALP activity in the osteoblasts-osteoclasts co-culture system.To observe the expression of Interleukin-11(IL-11) mRNA on osteoblast with the psoralen,based on the experimental results,we have discussed the mechanism of the psoralen in prevention and treatment of osteoporosis.Methods:1.Osteoblasts and osteoclasts were isolated respectively from SD rats 1 and 5 days old,the osteoblasts-osteoclasts co-culture system was built to prevent the two kinds of cells from contact and allow the media to exchange.2.The psoralen's various concentration on osteoblastic proliferation was assayed by MTT,which in order to determination the optimal concentration.3.The ALP activity of osteoblast was assayed with PNPP;the mineralization of OB was observed by chinalizarin-staining method.4.The expressed levels of IL-11 mRNA of osteoblast which were interfered in psoralen were assayed by RT-PCR.Result:1.Protein dispersing experiment:after detection by Coomassie Brilliant blue staining,we demonstrated that the protein in OB culture room dispersed to OC culture room through membrane within 8 hours,which suggested that the establishment of plane osteoblast and osteoclast co-culture system was successful.2.In the osteoblasts-osteoclasts co-culture system,proliferation of osteoblasts in co-culture OD was faster(P<0.05) than that in mono-culture OD.3.The ALP activity and the average number of mineralized nodes of OB that in the osteoblasts-osteoclasts co-culture system was higher than the OB cultured alone(P<0.05).4.When the psoralen's concentration was 5μmol/L,the OD value of OB was higher than other groups(P<0.05) and the ALP activity was higher than control group(P<0.05).5.The IL-11mRNA expressed level of OB in the co-culture system which were interfered in psoralen was higher than the control groups(P<0.05).Conclusions:1.Comparing to the mono-culture system,proliferation,differentiation and mineraliziation of osteoblasts were enhanced in the osteoblasts-osteoclasts co-culture system.2.The psoralen in 5μmol/L obviously stimulated the osteoblastic proliferation and boosted the ALP activity.3.The psoralen obviously improved the level of expression of IL-11mRNA.
