| Osteoporosis(OP)is a systemic metabolic bone disease caused by bone loss and changes in bone structure.It will increase the fragility of bones,make it easy to fracture,and seriously affect the quality of life of patients.China has the highest incidence of osteoporosis in the world,and with the aging of our population,osteoporosis patients are also increasing sharply.This has become one of the urgent problems in our country today.At the same time,OP also has many adverse effects on orthodontics.Compared with healthy orthodontic patients,orthodontic patients who also suffer from OP will experience more anchorage loss,uncontrolled tooth movement,root and alveolar bone resorption during orthodontic treatment.Seriously affect the orthodontic experience and orthodontic effect of OP patients.There are many pathogenic factors for OP,but its specific pathogenesis is still unclear.Traditional Chinese and Western medicine hold different views on it.The imbalance between osteoblast-mediated bone formation and osteoclast-mediated bone resorption is considered to be an important cause of the onset of OP,based on this western medicine for the treatment of OP to promote bone formation,inhibit bone resorption,There are three types of drugs.However,no specific medicine has been found to treat osteoporosis,and most western medicines used to treat osteoporosis have relatively large side effects.With the gradual improvement of my country’s international influence and the continuous deepening of my country’s emphasis on traditional Chinese medicine and research.At home and abroad,OP’s solutions are gradually converted to traditional Chinese medicine.Yishen Buguye(YSBGY)is a traditional Chinese medicine preparation composed of twelve kinds of herbs,which has been widely used clinically to strengthen muscles and bones in my country.Among them,the multi-flavored Chinese medicine has been proved to have anti-inflammatory,anti-oxidative stress,and regulate the physiological activities of osteoblasts and osteoclasts.And in the theoretical system of Chinese medicine,the pathogenesis of OP is divided into three types: kidney deficiency,spleen deficiency,and spleen and kidney deficiency,and YSBGY has nourishing liver,nourishing kidney,invigorating spleen,calming heart,replenishing qi,etc.Kind of role.Therefore,YSBGY can treat OP in both Chinese and Western medicine theory.In this study,we established an osteoporosis model by intraperitoneal injection of dexamethasone(Dex)in C57BL/6 mice,and studied the anti-osteoporosis effect of YSBGY by administering YSBGY to osteoporosis model mice.H&E staining and Giemsa staining on the internal organs and femurs of experimental mice found that YSBGY improved Dex-induced interstitial edema in the liver and kidney of OP mice,and increased the number of femoral trabecular bone and chondrocytes.Micro CT scan analysis of the femur and tibia of experimental mice found that YSBGY can increase the thickness of the bone cortex of Dex-induced OP mice,increase the bone density of the femur,and reverse the adverse bone structural changes in the femur and tibia of OP mice.Enzyme-linked immunoassay on peripheral blood serum of experimental mice found that YSBGY can increase serum type I collagen(COL-I),osteocalcin(BGP OCN),osteopontin(OPN),and osteogenic protein 2(BMP-2),type II osteogenic protein receptor protein(BMPR-2),type I collagen C terminal peptide(CTX-1),and reduce anti-tartrate phosphatase 5(trap5)and activated T cell nuclear factor 1recombinant protein(NFATc1)suggests that it can promote the differentiation of osteoblasts while inhibiting the differentiation of osteoclasts.In addition,the detection of inflammatory factors in the peripheral blood serum of experimental mice found that YSBGY can also up-regulate the expression level of interleukin 33(IL-33)and downregulate IL-1,IL-7 and tumor necrosis factor in OP mice.α(TNF-α)reduces inflammation in mice.Through the detection of oxidative stress factors in the peripheral blood serum of experimental mice,it was found that YSBGY enhanced the serum superoxide dismutase(SOD)and catalase(CAT)levels of OP mice,indicating that it has antioxidant properties.We used Western blot experiments to semi-quantitatively detect osteogenic and osteoclast-related proteins in the bone tissues of experimental mice,and found that YSBGY can make type I collagen(COL-I)and bone calcium in the bone tissues of OP mice.The expression levels of OCN(Osteocalcin),osteopontin(OPN),osteogenic protein 2(BMP-2)and Runx2 protein(Runx2)are increased,while inhibiting activated T cell nuclear factor 1 recombinant protein(NFATc1)and antitartrate phosphatase The expression of 5(trap5),which is consistent with the serum test results,proves that YSBGY can promote the differentiation of osteoblasts and inhibit the formation of osteoclasts.This study first confirmed the anti-osteoporosis effect of YSBGY on Dex-induced osteoporosis in C57BL/6 mice,and its mechanism of action may be related to its regulation of the physiological activity balance between osteoblasts and osteoclasts and the regulation of inflammatory factors.Related,this research provides experimental evidence for YSBGY as a drug to promote bone formation in clinic. |
