Objective To detect whether or not hepatitis B virus (HBV) DNA occurred in the ovarian tissue of the hepatitis B virus surface antigen (HBsAg)-positive patients with ovarian tumor, whether HBV could infect human ovarian tissue and germ cells causing vertical transmission. To construct and identify eukaryotic expression vector pcDNA3.1/CT-GFP-TOPO-HBS which carries HBV surface antigen gene (HBV S). Methods Molecular biology techniques, such as polymerase chain reaction (PCR), fluorescence in situ hybridization (FISH) and reverse transcription PCR(RT-PCR) were used to detect HBV DNA in the ovary paraffin sections of the patients with ovarian tumors, to construct recombination vector pcDNA3.1/CT-GFP-TOPO-HBS, and to detect HBV S gene expression in Hep G2. Results HBV DNA existed in the ovarian tissue cells of the HBsAg-positive patients with ovarian tumor and no HBV DNA was detected in the HBsAg-negative control group patients. Difference in the HBV DNA existence between these two groups was significant (P < 0.01 ). Constructed vector PCDNA3.1-N-GFP-TOPO-HBS.
