Objective To obverse the effect of rosiglitazone on no-reflow phenomenon and ventricle remodeling in rats undergoing acute coronary artery ischemic-reperfusion,and to explore the possible mechanism of the effect of rosiglitazone on no-reflow phenomenon and ventricle remodelingMethods 96 healthy Sprauge Dawley rats were randomly divided into acute effect group and chronic effect group,48 rats in one group,and every group was randomly divided into normal sub-group,sham operation sub-group,control sub-group and treatment sub-group (12 rats in every sub-group)。rats in normal sub-group and sham sub-group were given with 5ml normal saline by gavage for one week every morning,so done rats in control sub-group and treatment sub-group with rosiglitazone in 5ml normal saline。Coronary blood flow of control sub-group and treatment sub-group both were occluded for 90min followed by reperfusion。Normal sub-group rats were not operated,left anterior descending coronary was through by the line,after operation,acute observing sub-group and chronic effect group were housed for 24 hours and 4 weeks,respectively。observation index:①acute effect group: After four weeks,to calculated IVST,LVPWT,LVET and LVFS thought IVSd,IVSs,LVDd,LVDs,LVPWd,LVPWs,LVEDV and LVESV,which were observed by cardiac ultrasound。to assessed myocardial I/R area by Evans blue stains;the account of white cell in microscope by H-E stains,and the expression of ICAM-1 mRNA and P-selectin mRNA in myocardial I/R area by Semi-quantitative RT-PCR。②chronic effect group:to calculate LVSI and LVEI,LVEF and LVFS,which were observed by cardiac ultrasound,LVMI thought the weight of rat's body / left ventricular,and the extent of collagen with collagen stains。Resultsacute effect group(1) Index of left ventricular structure and function:there was no significant differentce of IVS and LVPW among four sub-groups(P>0.05);there was no significant difference of LVD,LVEDV,LVESV,LVPWT,IVST,LVFS and LVEF between normal sub-group and sham operation sub-group(P>0.05),LVD,LVEDV and LVESV of control sub-group and treatment sub-group were significantly more than normal sub-group and sham sub-group (P<0.05 or P<0.01),then LVPWT,IVST,LVFS and LVEF were significantly less than normal sub- group and sham sub-group(P<0.05 or P<0.01),LVD,LVEDV and LVESV of treatment sub-group were significantly less than control sub-group(P<0.05),LVPWT,IVST,LVFS and LVEF were significantly less than control sub-group(P<0.05)。(2) Myocardial no-reflow'area:no-reflow's area of treatment sub- group was significantly less than control sub-group(30.71±5.89%vs 35.94±6.53%,P<0.05)。(3) The account of white cell of I/R myocardium:there was no significant difference of the account of white cell of I/R myocardium between in normal sub-group and in sham operation sub-group(562.5±67.1对517.2±56.4,P>0.05);the account of white cell of I/R myocardium in control sub-group and treatment sub-group was both significantly more than normal sub-group and sham operation(P<0.01),then that in treatment sun-group was significantly less than that in control sub-group(1256.7±147.9 vs 1473.4±163.5,P<0.05)。(4) Expression of ICAM-1 and P-selictin mRNA in I/R myocardium:There was significant difference of the relative gray values of ICAM-1 mRNA in I/R myocardium of between normal sub-group and sham operation sub-group(18.4±1.9%vs 19.2±2.3%,P>0.05);the relative gray values of ICAM-1 mRNA in I/R myocardium of control sub-group and treatment sub-group were both significantly higher than normal sub-group and in sham operation sub-group(P<0.01),then that in treatment sub-group was significantly lower than control sub-group(61.4±3.8%vs 78.5±4.3%,P<0.05)。There was significant difference of the relative gray values of P-selictin mRNA in I/R myocardium between of normal sub-group and of sham operation sub-group(19.7±2.0%vs 20.3±1.9%,P>0.05);the relative gray values of P-selictin mRNA in I/R myocardium of control sub-group and treatment sub-group were both significantly higher than normal sub-group and in sham operation sub-group(P<0.01),then that in treatment sub-group was significantly lower than control sub-group(62.3±4.0%vs 77.2±3.9%,P<0.05)。chronic effect group(1) Index of left ventricular structure and function,there was all no significant difference of rats' body weight among four groups(P>0.05);there were all no significant difference of LVMI,IVS,LVEI,LVSI,LVEF and LVFS between in normal sub-group and in sham operation sub-group(P>0.05);LVMI,IVS and LVEI of control sub-group and treatment sub-group were all significantly more than these of normal sub-group and sham operation sub-group(P<0.05 or P<0.01),LVSI,LVEF and LVFS significantly less than normal sub-group and sham operation sub-group(P<0.05 or P<0.01),LVRM,LVSI,IVS and LVEI of treatment sub-group were all significantly less than control sub-group(P<0.05),then LVEF and LVFS significantly more than control sub-group(P<0.05)。(2) the extent of I/R myocardial collagen:There was no significant difference of the extent of I/R myocardial collagen between in normal sub-group and in sham operation sub-group(4.52±0.36%vs 4.87±0.25%,P>0.05);) the extent of I/R myocardial collagen of control sub-group and treatment sub-group were both significantly more than of normal sub-group and sham operation sub-group(P<0.01),then that of treatment sub-group significantly less than of control sub-group(30.03±4.51%vs38.24±3.78%,P<0.05=。Conclusion Rosiglitazone reduced myocardial no-reflow after actue myocardial ischemic-restoration coronary blood flow,inhibited choric ventricular remodeling,enhance acute and choric cardial function,which mechanism was related with inhibiting expression of ICAM-1 and P-selectin,reducing white cell aggreating ischemic area and diminishing collagen deposition of ischemic area。...
