Objective To detect the infection of Epstein-Barr virus,human cytomegalovirus and herpes simplex virus in tuberculosis and the level of soluble interleukin-2 receptor (sIL-2R) in serum after intensification treatment.Methods EBV specific DNA fragment BamH I-W was tested in peripheral blood monocytes of 56 pulmonary tuberculosis patients and 43 normal controls by polymerase chain reaction(PCR)-southern blotting.EBV-VCA-IgM,HCMV-IgM and HSV-IgM in serum were assessed by ELISA.The level of sIL-2R in serum was examined with ELISA.Results①Among 56 pulmonary tuberculosis patients 28 were EBV specific DNA fragment positive,The positive rate was 50%,while among 43 normal controls 2 were found,the positive rate was 4.65%.The positive rate of pulmonary tuberculosis patients was obviously higher than that of normal controls(χ~2=23.684,P<13.01).②The positive rate of EBV-VCA-IgM in 56 tuberculosis patients was 17.9%(10/56).The positive rate of EBV-VCA-IgM in 43 normal controls was 4.65%(2/43).There was markedly difference between patients and normal controls(χ~2=3.982,P<0.05)o The positive rate of HCMV-IgM in 56 pulmonary tuberculosis patients was 14.29%(8/56),HCMV-IgM wasn't detected in 43 normal controls.There was markedly difference by statistics analysis.③The level of sIL-2R in 42 pulmonary tuberculosis patients was 1087±841pg/ml before treatment.The level of sIL-2R was 751±120 pg/ml after 2 months treatment.The level of sIL-2R in normal controls was 214±66 pg/ml,There was significant difference between patients and normal controls(t'=6.198,24.053,P<0.01),The difference of the level of sIL-2R before and after the treatment was significant(t=2.563,P<0.05);the level of sIL-2R in the retreated patients before and after the treatment were 1542±698pg/ml and 962±284 pg/ml,markedly higher than those of the controls(t'=7.124,9.724,P<0.01), the difference of the contents of sIL-2R before and after the treatment was significant (t=2.900,P<0.01).⑥The level of sIL-2R in pulmonary tuberculosis patients with HCMV-IgM and HSV-IgM was 1688±754 pg/ml,while the level of sIL-2R of pulmonary tuberculosis patients without HCMV-IgM or HSV-IgM was 1006±738 pg/ml,there was significant difference between two groups(t=2.832,P<0.01 ).Conclusion①Compared with normal controls,there are higher EBV DNA positive rate in the peripheral blood mononuclear cells in pulmonary tuberculosis patients,This suggested that the reactivation of EBV might exist in pulmonary tuberculosis patients.②The active infection of HCMV or HSV existed in some pulmonary tuberculosis patients.③The level of sIL-2R in pulmonary tuberculosis patients with and without herpes virus was obviously difference,it suggested herpes virus active infection may affect the patients' immune function.④The changes of sIL-2R level in pulmonary tuberculosis patients before and after therapy can be used as an useful referential indicator for monitoring and predicting pulmonary tuberculosis.
